花椒叶多糖对小鼠运动疲劳作用及其机制研究

摘　　要：采用小鼠负重游泳模型来探究花椒叶多糖抗疲劳作用并初步研究其作用机制。采用超声波辅助法提取花椒叶得到花椒叶多糖,将其灌胃至小鼠,研究对小鼠负重游泳的影响及其抗疲劳机制。结果表明花椒叶多糖显著提高小鼠负重游泳时间,可以调节疲劳小鼠的生化指标(TTS、CTC、LD及BUN)来抵御疲劳,还可以调节肝糖原和肌糖原的储备维持血糖水平以缓解小鼠运动后疲劳。花椒叶多糖缓解小鼠运动疲劳的机制是通过提高具有清除和修复氧化自由基的抗氧化酶类(SOD、GSH-px和CAT)活性、抑制脂肪氧化酶和乳酸脱氢酶活性以阻碍MDA和乳酸生成来提高抗氧化活性,改善细胞抗氧化系统,又可以上调小鼠CYP11A1m RNA及CYP17A1 mRNA水平以调节TTS含量,以及通过调节NRF-1-TFAM通路提高线粒体抗氧化能力达到抗疲劳目的。花椒叶多糖是一种能有效缓解运动后疲劳的功能性食品成分。Using weight-bearing swimming model to explore the anti-fatigue effect of Zanthoxylum bungeanum leaf polysaccharide on mice as well as its mechanism.The polysaccharide from Zanthoxylum bungeanum leaves was extracted by ultrasonic assisted method,and was gavaged to weight-bearing swimming mice.The results showed that polysaccharide significantly increased the mice swimming time,its mechanism was by regulating biochemical indexes(TTS,CTC,LD and BUN)to resist fatigue,by regulating the liver and muscle glycogen to maintain blood glucose levels to alleviate post exercise fatigue of mice.The antioxidant system was improved by increasing the activities of antioxidant enzymes(SOD,GSH-px and CAT)that could scavenge and repair oxidative free radicals,inhibit the activity of lipoxygenase and lactate dehydrogenase.MDA and Lactic acid production were inhibited and antioxidant activity was increased,thus to improve cellular antioxidant system.It could also up-regulate the levels of CYP11A1 mRNA and CYP17A1 mRNA in mice to regulate the content of TTS and improve the antioxidant capacity of mitochondria by regulating NRF-1-TFAM pathway to achieve anti-fatigue.The polysaccharide from Zanthoxylum bungeanum leaves could be used as a food functional component to effectively alleviate fatigue after exercise.
