摘 要:为筛选高效降解豆粕菌株并探究其固态发酵最优发酵条件,从广西钦州红树林根部土壤中筛菌,经形态学观察、革兰氏染色、16S测序分析和生理生化鉴定该菌株。以不同碳源、料水比、接种量、发酵时间及发酵温度为影响因素,进行单因素优化及L16(45)正交试验。并探究该条件下豆粕中抗营养因子含量。结果表明:筛选到一株高效降解豆粕的菌株S7,经鉴定为乳酸肠球菌(Enterococcus lactis)。在以葡萄糖为碳源,料水比1:1,接种量8%,37℃发酵36 h时酸溶性蛋白含量最高,为12.19%。在此条件下发酵豆粕时,豆粕中的大豆β-伴球蛋白含量降低了60.7%,大豆球蛋白含量降低了75.5%,脲酶活性降低了60.3%,胰蛋白酶抑制因子含量降低了72.1%。该研究结果为进一步提高豆粕利用率及营养价值提供理论支持。In order to screen the strains with high degradation efficiency of soybean meal and explore the optimum fermentation conditions of its solid-state fermentation.Screen bacteria from mangrove root soil in Qinzhou,Guangxi were identified by morphological observation,Gram staining 16S sequencing analysis and physiological and biochemical identification.Single factor optimization and L16(45)orthogonal test were carried out with different carbon sources,feed-water ratio,inoculation amount,fermentation time and fermentation temperature as influencing factors.The content of anti-nutritional factors in soybean meal under these conditions was investigated.The results showed that a strain S7 was identified as Enterococcus lactic.When glucose was used as carbon source,the ratio of feed to water was 1:1,and the inoculation amount was 8%and fermented at 37℃for 36 h,the highest content of acid soluble protein was 12.19%.Under these conditions,the content of soybeanβ-conglobulin decreased by 60.7%,the content of soybean globulin decreased by 75.5%,the activity of urease decreased by 60.3%,and the content of trypsin inhibitor decreased by 72.1%.The results of this study provide theoretical support for further improving the utilization rate and nutritional value of soybean meal.
