摘 要:目的:构建大鼠口腔黏膜炎模型评价并比较复方苦参注射液(Compound Kushen Injection,CKI)不同给药方式对放射性口腔黏膜炎的的治疗作用。方法:选取检疫合格SPF级SD大鼠90只,雌雄各半,体重178.6~388.3 g,按体重与性别随机分为9组,分别为正常对照组、模型对照组、金因肽组、复方苦参注射液静脉组(1.8 mL/kg)、复方苦参注射液喷雾组、口服复方苦参注射液剂量1、2、3、4组(0.45、0.9、1.8、3.6 mL/kg),每组10只/组。按15 Gy剂量采用X射线照射大鼠口部及内侧口腔黏膜,另取10只正常对照组不做处理,照射后每只动物灌胃给予4 mL/只0.9%氯化钠注射液。各组动物分别于造模后24 h给予不同浓度的药物,每天对各组动物进行一般临床观察,定期监测体重、摄食量及口腔黏膜评分,末次给药后各组大鼠安乐死,取口腔黏膜组织进行细胞因子IL-6、TNF-α、IL-1β的含量检测,以及口腔黏膜组织进行组织病理学检查。结果:复方苦参注射液口服剂量各剂量均能减轻X射线致大鼠口腔黏膜病变程度,其中1.8、3.6 mL/kg剂量的复方苦参注射液能明显增加动物体重增长率和摄食量,降低口腔黏膜评分,并能降低IL-6、IL-1β水平。复方苦参注射液口服给药对X射线致大鼠口腔黏膜具有明显保护作用,在同等剂量下,1.8 mL/kg浓度的复方苦参注射液治疗大鼠口腔黏膜炎的效果优于复方苦参注射液静脉给药。金因肽和复方苦参注射液静脉给药均能明显增加动物体重和摄食量,在第四天与第七天能明显降低口腔黏膜评分,同时降低了口腔黏膜组织中IL-6和IL-1β的水平。结论:复方苦参注射液口服给药对X射线致大鼠口腔黏膜炎具有明显保护作用,其作用机制可能与降低炎性因子水平有关。在同等剂量下,复方苦参注射液口服剂量(1.8 mL/kg)治疗大鼠口腔黏膜炎的效果优于复方苦参注射液静脉给药。Objective:To construct a rat oral mucositis model to evaluate and compare the therapeutic effects of different administration methods of Compound Kushen Injection(CKI)on radiation oral mucositis.Methods:A total of 90 SPF-grade SD rats that were quarantine-qualified were selected,half of male and female,weighing 178.6~388.3 g,randomly divided into 9 groups according to body weight and gender.They were normal control group,model control group,Jinyin peptide(recombinant human epidermal growth factor)group.CKI intravenous injection group(1.8 mL/kg),CKI spray group,CKI oral dose 1,2,3,4 groups(0.45,0.9,1.8,3.6 mL/kg),10 rats for each group.The mouth and inner oral mucosa of rats were irradiated with X-rays at a dose of 15 Gy,and another 10 normal control groups were taken without treatment.After irradiation,each animal was injected 4 mL/mouse of 0.9%sodium chloride.Animals in each group were given different concentrations of drugs at 24 hours after modeling.General clinical observations were performed on each group of animals every day.Body weight,food intake and oral mucosal scores were regularly monitored.After the last administration,rats in each group were euthaniz ed and oral mucosal tissues were taken.Carry out cytokine IL-6,TNF-α,IL-1βcontent detection,and oral mucosal tissue for histopathological examination.Results:All oral doses of CKI can alleviate the degree of X-ray-induced oral mucosal lesions in rats.Among them,CKI at the doses of 1.8 and 3.6 mL/kg can significantly increase the growth rate of animal weight and food intake,and reduce oral mucosal score,and can reduce IL-6,IL-1βlevels.Oral administration of CKI has a significant protective effect on the oral mucosa of rats caused by X-rays.At the same dose,the CKI oral group 1.8 mL/kg was more effective than CKI intravenous injection group(1.8 mL/kg)in rats.Jinyin peptide and CKI intravenous injection group can significantly increase animal body weight and food intake,reduce oral mucosal scores on the fourth and seventh days and reduce IL-6 and IL-1β
