摘  要:目的:通过观察乳腺癌组织中泛素特异性蛋白酶20(USP20)、Ezrin表达变化及其与临床病理特征之间的关系,探讨乳腺癌组织中USP20与Ezrin表达的相关性及临床意义。方法:免疫组织化学及免疫荧光方法用于检测USP20与Ezrin阳性细胞的表达变化以及细胞定位,并分析它们与乳腺癌患者临床病理特征的关系;shRNA-USP20慢病毒转染乳腺癌MDA-MB-231细胞,通过荧光定量PCR与Western Blot检测USP20基因及蛋白的过表达水平,然后观察过表达USP20后Ezrin基因和蛋白的表达变化。结果:USP20与Ezrin在乳腺癌组织中的阳性表达率分别为69.6%和78.5%,而在邻近相对正常组织中的表达均为阴性,与邻近相对正常组织相比,乳腺癌组织中USP20与Ezrin阳性表达均显著增加,差异均具有统计学意义(P<0.01);USP20与Ezrin阳性表达均主要在乳腺癌组织细胞质中,少量在细胞核。两者在乳腺癌组织中的阳性表达与患者的年龄、性别、WHO分级、分子分型和有无淋巴结转移均无关;shRNA-USP20慢病毒转染乳腺癌MDA-MB-231细胞后,USP20蛋白及mRNA的表达均明显增加(P<0.01),过表达USP20后Ezrin的mRNA表达不变,蛋白表达显著增加(P<0.01)。结论:乳腺癌组织中USP20和Ezrin表达呈正相关关系,且过表达USP20后Ezrin蛋白表达显著增加,USP20和Ezrin可能共同促进乳腺癌发生发展,其机制可能与USP20去泛素化作用有密切关系,这为临床治疗、预后及进一步研究提供重要的实验依据。Objective:To investigate USP20 and Ezrin expression in breast cancer and its relationship with clinicopathological features,and to find out the correlation of USP20 and Ezrin and the clinical significance.Methods:Immunohistochemistry and double immunofluorescence histochemistry were used to observe the expression changes and cell localization of positive cells of USP20 and Ezrin,and to analyzeits relationship with clinicopathological features.After shRNA-USP20 lentivirus was transfected into breast cancer MDA-MB-231 cells,and USP20 gene and protein was detected by fluorescence quantitative PCR and Western Blot,then the expression of Ezrin was observed after USP20 overexpression.Results:The positive expression rates of USP20 and Ezrin were 69.6%and 78.5%in breast cancer tissues,while they were negatively expressed in the adjacent normal tissues,and the differences were statistically significant(P<0.01).The positive expressions of USP20 and Ezrin were mainly in the cytoplasm of breast cancer tissues,and a small amount in the nucleus.Their positive expression in breast cancer tissues was not correlated with age,gender,WHO grading,molecular typing,or lymph node metastasis.After shRNA-USP20 lentivirus was transfected into breast cancer MDA-MB-231 cells,the expression of USP20 protein and gene was significantly increased(P<0.01),and after USP20 overexpression,there was no change of Ezrin mRNA,and Ezrin protein expression was significantly increased(P<0.01).Conclusion:USP20 expression was positively correlated with Ezrin expression in breast cancer significantly,and Ezrin protein expression was significantly increased after USP20 overexpression.So,USP20 and Ezrin may work together to promote breast cancer development.The mechanism may be closely related to the deubiquitination of USP20,which may provide important experimental basis for clinical treatment,prognosis and further research.
