摘 要:目的:研究HER-2表达对胃癌细胞侵袭、迁移及黏附的影响,并探讨HER-2表达与EMT相关蛋白的关系。方法:利用小干扰RNA技术,在MKN-45胃癌细胞中沉默HER-2表达;采用实时荧光定量PCR法及Western blot法验证HER-2沉默效果,并将实验分为空白对照组、阴性对照组和HER-2沉默组;采用实时荧光定量PCR法及Western blot法检测各组细胞EMT相关蛋白E-cadherin、Vimentin、Snail的mRNA和蛋白表达情况。采用Transwell小室技术检测细胞侵袭及迁移能力;MTT实验检测细胞黏附能力。结果:沉默MKN-45胃癌细胞株中HER-2表达后,HER-2沉默组Vimentin、Snail表达水平以及侵袭、迁移及黏附能力明显低于空白对照组和阴性对照组;HER-2沉默组E-cadherin表达水平明显高于空白对照组和阴性对照组,差异均具有统计学意义(P<0.05)。结论:在胃癌细胞中,HER-2可能通过影响EMT相关蛋白表达从而增强胃癌细胞的侵袭、迁移及黏附能力。Objective:To study the effect of HER-2 expression on gastric cancer cell invasion,migration and adhesion,and to investigate the relationship between HER-2 expression and EMT related protein.Methods:HER-2 expression was silenced in MKN-45 gastric cancer cells by small interfering RNA.The silencing of HER-2 was verified by RT-PCR and Western blot,and the experiment was divided into blank control,negative control and HER-2 silencing groups.The mRNA and protein expression of E-cadherin,Vimentin and Snail(EMT related protein)were detected by RT-PCR and Western blot.Cell invasion and migration were measured by Transwell chamber technique.Cell adhesion were measured by MTT experiment.Results:After silencing of HER-2 expression in MKN-45 gastric cancer cell lines,Vimentin,Snail and adhesion of HER-2 silencing group were significantly lower than the blank control and negative control group.E-cadherin in HER-2 silencing group was significantly higher than the blank control and negative control group,and the differences were statistically significant(P<0.05).Conclusion:In gastric cancer cells,HER-2 may enhance the invasion,migration and adhesion ability of gastric cancer cells by affecting the EMT related proteins.
