结直肠癌患者microRNA-101、microRNA-182表达与病理特征及预后的关系

摘　　要：目的探讨结直肠癌组织中microRNA-101(miR-101)、microRNA-182(miR-182)表达与临床病理特征及预后的关系。方法选取2015年4月—2018年1月青海省人民医院收治的83例拟行结直肠癌根治术患者,术后随访5年。采用实时荧光定量聚合酶链反应(qRT-PCR)检测所有患者癌组织及癌旁组织miR-101、miR-182的表达;分析不同临床病理特征结直肠癌患者癌组织miR-101、miR-182的表达;采用多因素Cox比例风险模型分析影响结直肠癌患者术后复发转移的因素;分析结直肠癌患者癌组织miR-101、miR-182表达与术后复发转移的关系。结果癌组织miR-101 mRNA相对表达量低于癌旁组织,miR-182 mRNA相对表达量高于癌旁组织(P<0.05);低分化、病理分期Ⅲ期、有浆膜浸润、有淋巴结转移的结直肠癌患者癌组织miR-101mRNA相对表达量低于中高分化、病理分期Ⅱ期、无浆膜浸润、无淋巴结转移者(P<0.05),miR-182 mRNA相对表达量的比较结果则相反(P<0.05);多因素Cox比例风险模型分析结果显示,病理分期[RR(95%CI:3.001,17.726)、分化程度[RR(95%CI:2.119,12.516)]、miR-101[RR(95%CI:1.842,10.881)]、miR-182[RR(95%CI:1.850,10.925)]是影响结直肠癌患者术后复发转移的因素(P<0.05);miR-101高表达患者的无病生存率高于低表达患者(P<0.05),miR-182低表达患者的无病生存率高于高表达患者(P<0.05)。结论结直肠癌患者癌组织miR-101、miR-182的表达与临床病理特征及预后有关,miR-101低表达、miR-182高表达患者治疗后复发转移风险高。Objective To investigate the relationship of tissue miR-101 and miR-182 levels with clinicopathologic features and prognosis in patients with colorectal cancer.Methods Eight three patients who were admitted to the hospital from April 2015 to January 2018 for radical resection of colorectal cancer were selected and followed up for 5 years.The relative expression levels of miR-101 and miR-182 in cancer tissues and adjacent tissues of all patients were detected.The relative expression levels of miR-101 and miR-182 in cancer tissues and adjacent tissues were compared.The expressions of miR-101 and miR-182 in cancer tissues of colorectal cancer patients with different clinicopathological characteristics were analyzed.Factors influencing postoperative recurrence and metastasis in patients with colorectal cancer were analyzed.The relationship between the expression of miR-101 and miR-182 and postoperative recurrence and metastasis in cancer tissues of colorectal cancer patients was analyzed.Results The relative expression level of miR-101 in cancer tissue was lower than that in paracancer tissue,and that of miR-182 was higher than that in paracancer tissue(P<0.05).The relative expression level of miR 101 in cancer tissues of colorectal cancer patients with low differentiation,stageⅢ,serous invasion,and lymph node metastasis was lower than that of patients with medium and high differentiation,stageⅡ,no serous invasion,and lymph node metastasis,respectively(P<0.05).The relative expression level of miR-182 in cancer tissues of colorectal cancer patients with low differentiation,stageⅢ,serous invasion,and lymph node metastasis was higher than that of patients with medium and high differentiation,stageⅡ,no serous invasion,and lymph node metastasis,respectively(P<0.05).Cox regression analysis showed:pathological stage[RR=7.294(95%CI:3.001,17.726)],differentiation degree[RR=5.150(95%CI:2.119,12.516)],relative expression level of miR-101[RR=4.477(95%CI:1.842,10.881)],and relative expression of miR-182[RR=4.495(95%CI:1.8
