摘  要:试验旨在提高蛋白核小球藻(Chlorella pyrenoidosa)的生物量和蛋白含量,对发酵配方和发酵工艺进行优化。采用Design-Expert 12 Perfect软件对复合碳源、氮源和维生素等13种因子进行Plackett-Burman筛选,得到显著效应因子为复合碳源(葡萄糖+淀粉,按2.5∶1.0的比例添加能够增加核蛋白小球藻发酵产量)、有机氮源(尿素)、维生素B_(12)(VB_(12))。对上述3个显著效应因子进行Box-Behnken试验设计,通过响应面分析获得最优添加量为复合碳源7.4 g/L、尿素8.8 g/L、VB_(12) 0.4 g/L。经250 mL和1 000 mL三角摇瓶培养基试验验证以及50 L发酵罐补料工艺验证,得出3个不同水平发酵的效果均高于对照组,藻体密度分别提高38.9%、31.1%、42.5%,蛋白浓度分别提高29.8%、31.0%、31.4%。研究表明,优化补料条件下,蛋白核小球藻50 L发酵罐OD_(680 nm)值为0.929,干重91.64 g/L,蛋白含量达43%。The study was to increase the biomass and protein content of Chlorella pyrenoidosa,the fermentation recipe and fermentation conditions were optimized.Using Design-Expert 12 Perfect software,Plackett-Burman screening was performed on 13 factors including complex carbon source,nitrogen source and vitamins,the significant effect factors were found to be complex carbon source(glucose + starch,the fermentation yield of Chlorella pyrenoidosa could be increased by adding them at a ratio of 2.5∶1.0),organic nitrogen source(urea),and VB_(12).The Box-Behnken experimental design was carried out using the above three significant effect factors,and the optimized process was obtained by response surface analysis,which were composite carbon source 7.4 g/L,urea 8.8 g/L and VB_(12) addition 0.4 g/L.The results of 250 mL and 1 000 mL triangle flask medium test and 50 L fermenter feeding process verification showed that the final three different levels of fermentation were higher than the control group,the algal density increased by 38.9%,31.1% and 42.5%,and the protein concentration increased by 29.8%,31.0% and 31.4%,respectively.Under the optimized feeding conditions,the OD_(680 nm) of the 50 L fermentor of Chlorella pyrenoidosa was 0.929,the dry weight was 91.64 g/L,and the protein content was 43%.
