全自动凯氏定氮仪在综合实验教学中的应用

摘　　要：一种测量食品内蛋白质含量的凯氏定氮法用于实验教学,其目的是使学生掌握全自动凯氏定氮仪的测定原理及操作技能。该实验涉及溶液配制、分析滴定、石墨消解仪操作、全自动凯式定氮仪的调试、莜面样品测试和仪器的清洗维护。本文介绍了实验所用仪器、药品试剂和测试的样品;石墨消解仪消化操作步骤;凯氏定氮仪测定原理、仪器调试及样品测试,实验对两种消化方案、消化效果和莜面含蛋白质量的测定结果进行了初步对比分析。该实验已在两届学生中实施,提升了学生的综合实验实践技能。In order to have a better understanding of the determination principle and master the automatic Kjeldahl apparatus operation skills,a comprehensive experiment on the determination of total protein content in oat noodles(Kjeldahl method)is applied in experimental teaching.The experimental process covers solution preparation,titration,graphite digestion apparatus operation,automatic Kjeldahl apparatus commission,oat noodles samples test and washing maintenance of the automatic Kjeldahl apparatus.The experimental apparatus,reagents,and the samples to be tested involved in the experiment were introduced.Digestion procedures of the graphite digestion apparatus were illustrated.The determination principle,apparatus adjustment,and samples tests via automatic Kjeldahl apparatus were expounded.Besides,two different digestion schemes,the digestion effects,the digestion results,and protein content results were discussed and compared preliminarily.This experiment has been carried out for two sessions,which improve the undergraduates’comprehensive practical ability.
