沉默lncRNA ST7-AS1通过Wnt/β-catenin信号通路对肝癌细胞增殖、迁移和侵袭影响的实验研究

摘　　要：目的:探讨沉默长链非编码RNA(lncRNA)致癌性抑制基因7反义RNA1(ST7-AS1)通过Wnt/β-连环蛋白(β-catenin)信号通路对肝癌细胞增殖、迁移和侵袭的影响。方法:肝癌细胞系HCCLM3分成Control组、Vector组、ST7-AS1组、sh-NC组、sh-ST7-AS1组和sh-ST7-AS1+Wnt/β-catenin通路激动剂(SKL2001)组。结果:与Control组比较,ST7-AS1组HCCLM3细胞的吸光度(A570)值及N-钙黏蛋白(N-cadherin)、波形蛋白(Vemintin)、Wnt1和β-catenin的表达升高,克隆数量和迁移、侵袭数目增多,上皮细胞钙黏蛋白(E-cadherin)和GSK-3β的表达降低(P<0.05)。Wnt/β-catenin通路激动剂SKL2001可部分逆转沉默ST7-AS1对HCCLM3细胞增殖、迁移、侵袭的影响(P<0.05)。结论:沉默ST7-AS1可通过抑制Wnt/β-catenin信号通路激活抑制肝癌细胞增殖、迁移和侵袭。Objective:To investigate the impacts of silencing long non-coding RNA(lncRNA)suppressor of tumorigenicity 7 antisense RNA 1(ST7-AS1)on the proliferation,migration and invasion of hepatoma cells through Wnt/β-catenin(β-catenin)signaling pathway.Methods:The liver cancer cell line HCCLM3 was divided into Control group,Vector group,ST7-AS1 group,sh-NC group,sh-ST7-AS1 group and sh-ST7-AS1+Wnt/β-catenin pathway agonist(SKL2001)group.Results:Compared with the control group,the A570 value and the expressions of N-cadherin,Vimentin,Wnt1 andβ-catenin of HCCLM3 cells in the ST7-AS1 group increased,the numbers of clones,migration and invasion increased,and the expressions of epithelial cell cadherin(E-cadherin)and GSK-3βdecreased(P<0.05).The Wnt/β-catenin pathway agonist SKL2001 could partially reverse the impacts of silencing ST7-AS1 on the proliferation,migration and invasion of HCCLM3 cells(P<0.05).Conclusion:Silencing ST7-AS1 can inhibit the proliferation,migration and invasion of liver cancer cells by inhibiting the activation of Wnt/β-catenin signaling pathway.
