摘  要:目的了解烟台地区非O1/O139群霍乱弧菌致病性现状。方法分别于2020年7月和11月、2021年的1月和4月对烟台市某两条河入海口采集样本32份,获得22株非O1/O139群霍乱弧菌分离菌株,采用脉冲场凝胶电泳(PFGE)对分离菌株进行分子分型,采用聚合酶链式反应(PCR)方法对该22株分离菌株的毒力基因ctxA、ctxB、ace、cep、zot、rtxA、hlyA、toxR、tcpA和cri进行检测分析。结果22株非O1/O139群霍乱弧菌菌株间的条带相似度为23.0%~100.0%,分为17个PFGE型,呈高度的多态性带型分布。22株非O1/O139群霍乱弧菌分离株主要携带3种毒力基因(rtxA、tcpA和zot),其中携带rtxA和tcpA最多,均为17株,其次是zot,共14株。14株菌株同时携带rtxA、tcpA和zot,5株不携带上述毒力基因。结论烟台地区入海口流行的非O1/O139群霍乱弧菌株污染情况多样化,主要携带具有黏附、毒素以及损伤小肠表皮细胞等方面的致病基因。Objective To understand the pathogenic characteristics of non-O1/O139 Vibrio cholera in Yantai.Methods A total of 32 samples were collected from the estuary of 2 rivers in Yantai in July and November 2020,and in January and April 2021.Among these samples,22 strains of non-O1/O139 Vibrio cholera were isolated and molecular typed by pulse field gel electrophoresis(PFGE).Their virulence genes including ctxA,ctxB,ace,cep,zot,rtxA,hlyA,toxR,tcp and cri were detected by polymerase chain reaction(PCR).Results The results showed that 22 strains of non-O1/O139 Vibrio cholerae were divided into 17 PFGE types with highly polymorphic distribution,and the similarities were between 23.0%and 100.0%.These isolates mainly carried 3 virulence genes(rtxA,tcpA and zot)with the highest positive rates of rtxA and tcpA(17 strains,respectively),followed by zot(14 strains).A total of 14 strains carried multiple genes of rtxA,tcpA and zot,while 5 strains did not carry these virulence genes.Conclusions The distribution of non-O1/O139 Vibrio cholerae in the estuary in Yantai is diverse.The pathogenic genes have the effect of adhesion,toxins,as well as the injury of small intestine epidermis cells.
