摘  要:[目的]为有效监控均质效果,研究一种快速有效方法对超高灭菌温乳中脂肪球粒径进行检测。[方法]利用8种染色剂设计试验步骤,通过制片、染色、镜检观察,开展样品检测,收集分析检测数据。[结果]不同染色剂染色脂肪球的现象存在差异。使用甲基橙染色法时指示剂和牛奶样品能有效融合,镜检时脂肪球染色效果好,背景干扰杂质少。使用另外7种指示剂染色时,指示剂和牛奶样品都不能有效融合,产生肉眼可见颗粒物质,镜检时会发现染色效果差,背景干扰杂质多。[结论]甲基橙染色法准确快速有效,可实现对均质化过程中脂肪球粒径的有效监控。[Objective]In order to effectively monitor the homogenization effect,a rapid and effective method was developed to detect the size of fat globules in ultra-high temperature sterilized milk.[Method]Test procedures were designed with 8 dyes,sample testings were carried out,and test data were collected and analyzed.[Result]The phenomenon of fat globules size stained by different dyes was different.When using methyl orange staining method,the indicator and milk sample can be effectively fused,and the fat globules staining effect was good and the background interference impurities were less.When the other 7 indicators were used for staining,the indicators and milk samples could not be effectively fused,resulting in visible solid matter,poor staining effect and more background interference impurities were found during microscopic examination.[Conclusion]Through comparative analysis of experiments,methyl orange staining is an accurate,rapid and effective method,which can realize effective monitoring of fat globule size in the homogenization process.
