上皮性卵巢癌组织中miR-515-5p、CSPG4的表达及临床意义

摘　　要：目的探讨上皮性卵巢癌(EOC)组织中微小RNA-515-5p(miR-515-5p)、硫酸软骨素蛋白聚糖4(CSPG4)的表达及临床意义。方法选取84例EOC患者,采用RT-qPCR法检测癌组织与癌旁组织中miR-515-5p、CSPG4mRNA表达。采用Pearson相关系数分析EOC组织中miR-515-5p与CSPG4 mRNA表达的相关性。根据EOC组织中miR-515-5p、CSPG4 mRNA表达均值分为高、低表达者,采用Kaplan-Meier法绘制不同miR-515-5p、CSPG4 mRNA表达EOC患者的生存曲线。结果与癌旁组织相比,卵巢癌组织中miR-515-5p表达低,CSPG4 mRNA表达高(P均<0.05)。经TargetScan数据库预测miR-515-5p与CSPG4的3’-非翻译区210~216处存在结合位点,Pearson相关系数分析显示,EOC组织中miR-515-5p与CSPG4 mRNA表达呈负相关(r=-0.710,P<0.05)。不同分化程度、国际妇产科联盟(FIGO)分期、淋巴结转移EOC组织中miR-515-5p、CSPG4 mRNA表达比较,差异均有统计学意义(P均<0.05)。随访3年,84例EOC患者3年累积生存率为57.14%(48/84)。K-M生存曲线分析显示,miR-515-5p高表达者累积生存率高于低表达者,CSPG4 mRNA高表达者累积生存率低于低表达者(P均<0.05)。结论EOC组织中miR-515-5p低表达和CSPG4 mRNA高表达,二者表达与分化程度、FIGO分期、淋巴结转移和预后有关;检测二者表达有助于判断EOC患者病情和预后评估。Objective To investigate the expression and clinical significance of microRNA-515-5p(miR-515-5p)and chondroitin sulfate proteoglycan 4(CSPG4)in epithelial ovarian cancer(EOC)tissues.Methods Eighty-four patients with EOC were selected,and miR-515-5p and CSPG4 mRNA expression levels in the cancer and para-carcinoma tissues were detected by RT-qPCR.Pearson correlation coefficient was used to analyze the correlation between miR-515-5p and CSPG4 mRNA expression in the EOC tissues.According to the mean values of miR-515-5p and CSPG4 mRNA expression in EOC tissues,they were divided into the high and low expression groups,and the survival curves of EOC patients with different miR-515-5p and CSPG4 mRNA expression levels were plotted by the Kaplan-Meier method.Results Compared with the para-carcinoma tissues,the miR-515-5p expression was lower and CSPG4 mRNA expression was higher in the ovarian cancer tissues(both P<0.05).TargetScan database predicted that miR-515-5p had binding sites at 210-216 of the 3'-untranslated region of CSPG4.Pearson correlation coefficient analysis showed that miR-515-5p was negatively correlated with CSPG4 mRNA expression in the EOC tissues(r=-0.710,P<0.05).Statistically significant differences were found in miR-515-5p and CSPG4 mRNA expression between EOC tissues with different degrees of differentiation,International Federation of Gynecology and Obstetrics(FIGO)stages,and lymph node metastases(all P<0.05).At 3 years follow-up,the 3-year cumulative survival rate of 84 EOC patients was 57.14%(48/84).Kaplan-Meier survival curve analysis showed that the cumulative survival rate was higher in the high miR-515-5p expression group than in the low expression group,and the cumulative survival rate was lower in the high CSPG4 mRNA expression group than in the low expression group(both P<0.05).Conclusions The miR-515-5p is low expressed and CSPG4 mRNA is highly expressed in EOC tissues,and both of them are related to differentiation degree,FIGO stage,lymph node metastasis and prognosis.Detection of their
