摘 要:目的探讨卵巢癌组织中微小RNA-581(miR-581)、叉头框转录因子O1(FOXO1)的表达及与临床病理特征和预后的关系。方法选取97例卵巢癌患者,采用RT-qPCR法检测癌组织与癌旁组织中miR-581、FOXO1mRNA表达。采用Pearson相关系数分析卵巢癌组织中miR-581与FOXO1 mRNA表达的相关性。根据卵巢癌组织中miR-581、FOXO1 mRNA表达均值分为高、低表达,采用Kaplan-Meier法绘制不同miR-581、FOXO1 mRNA表达卵巢癌患者生存曲线。结果与癌旁组织比较,卵巢癌组织中miR-581表达高,FOXO1 mRNA表达低(P均<0.05)。Pearson相关系数分析显示,卵巢癌组织中miR-581与FOXO1 mRNA表达呈负相关(r=-0.728,P<0.05)。不同分化程度、国际妇产科联盟(FIGO)分期、淋巴结转移卵巢癌组织中miR-581、FOXO1 mRNA表达比较,差异均有统计学意义(P均<0.05)。随访3年,死亡42例,3年累积生存率为56.70%(55/97)。K-M生存曲线分析显示,miR-581高表达者累积生存率低于miR-581低表达者,FOXO1 mRNA高表达者累积生存率低于FOXO1 mRNA低表达者(P均<0.05)。结论卵巢癌组织中miR-581高表达和FOXO1 mRNA低表达,二者表达与分化程度、FIGO分期、淋巴结转移和预后有关。Objective To investigate the expression levels of microRNA-581(miR-581)and forkhead box O1(FOXO1)in the ovarian cancer tissues and their relationships with clinicopathologic features and prognosis.Methods Ninety-seven ovarian cancer patients were selected.RT-qPCR was performed to detect miR-581 and FOXO1 mRNA expression in the cancer and para-carcinoma tissues.Pearson correlation coefficient was used to analyze the correlation between miR-581 and FOXO1 mRNA expression in ovarian cancer tissues.According to the mean values of miR-581 and FOXO1 mRNA expression in the ovarian cancer tissues,they were divided into the high and low expression groups,and the survival curves of ovarian cancer patients with different miR-581 and FOXO1 mRNA expression levels were plotted by the Kaplan-Meier method.Results Compared with para-carcinoma tissues,the expression of miR-581 was higher and the expression of FOXO1 mRNA was lower in the the ovarian cancer tissues(both P<0.05).Pearson correlation coefficient analysis showed that miR-581 was negatively correlated with FOXO1 expression in the ovarian cancer tissues(r=-0.728,P<0.05).Statistically significant differences were found in the miR-581 and FOXO1 mRNA expression between ovarian cancer tissues with different degrees of differentiation,International Federation of Gynecology and Obstetrics(FIGO)stages,and lymph node metastasis(all P<0.05).At 3 years of follow-up,there were 42 cases of deaths,with a 3-year cumulative survival rate of 56.70%(55/97).K-M survival curve analysis showed that the cumulative survival rate of the high miR-581 expression group was lower than that of the low miR-581 expression group,and the cumulative survival rate of the high FOXO1 mRNA expression group was lower than that of the low FOXO1 mRNA expression group(both P<0.05).Conclusion MiR-581 is highly expressed and FOXO1 mRNA is low expressed in the ovarian cancer tissues,which are related to the degree of differentiation,FIGO stage,lymph node metastasis and prognosis.
