摘  要:目的 观察“原络通经”针法对SAMP8小鼠认知障碍及突触相关蛋白表达的影响,探究“原络通经”针法对阿尔茨海默病(Alzheimer’s disease,AD)诱导的小鼠认知障碍的作用机制。方法 选取6只SAMR1同品系小鼠作为SAMR1正常对照组;另选取18只SAMP8小鼠,采用随机数字表法分为SAMP8空白组、SAMP8+“原络通经”针刺组和SAMP8+百会针刺组,每组各6只。其中SAMP8空白组每日定时抓取绑定不针刺,SAMP8+“原络通经”组针刺丰隆、太白、太溪、飞扬及百会穴,SAMP8+百会穴组仅针刺百会穴。各组针刺1次/d,干预30 d后,采用Morris水迷宫和旷场实验检测小鼠认知与空间学习记忆能力;HE染色观察小鼠海马CA1区神经元病理损伤程度;TUNEL检测小鼠海马组织神经元凋亡;免疫组织化学法检测小鼠海马组织突触素(SYN)和突触后膜蛋白-95(PSD-95)的表达;ELISA检测海马组织中β-淀粉样蛋白40(Aβ_(40))、磷酸化tau(p-tau)含量。结果 (1)与SAMP1组比较,其余3组小鼠逃逸路径、逃避潜伏期延长,第2象限停留的时间缩短,通过原平台位置的次数和进入旷场中心区域次数减少,差异均有统计学意义(P<0.05);与SAMP8空白组比较,“原络通经”组与百会穴组小鼠逃避潜伏期均缩短,第2象限停留的时间延长,通过原平台位置次数和进入旷场中心区域次数增加,且“原络通经”组小鼠改善情况优于百会穴组,差异均有统计学意义(P<0.05)。(2)各组小鼠海马组织HE与TUNEL染色显示,SAMR1组小鼠神经细胞形态、结构正常,排列有序,凋亡细胞数量少;与SAMR1组相比,SAMP8空白组小鼠神经细胞出现皱缩,数目减少,形态、排列不规则,空泡增加,凋亡细胞数量明显增多;与SAMP8空白组相比,“原络通经”组与百会穴组小鼠病理形态明显改善,凋亡细胞数量减少,且“原络通经”组优于百会穴组,差异具有统计学意义(P<0.05)。(3)与SAMR1组比较,SAMP8空白组小鼠海马组�Objective To observe the effects of“Yuanluo Tongjing”acupuncture on cognitive impairment and synaptic related protein expression in SAMP8 mice,and to explore the mechanism of“Yuanluo Tongjing”acupuncture on cognitive impairment induced by Alzheimer's disease(AD)in mice.Methods Six SAMR1 mice of the same strain were selected as normal control group.Another 18 SAMP8 mice were selected and divided into SAMP8 blank group,SAMP8+Yuanluo Tongjing acupuncture group and SAMP8+Baihui acupuncture group by random number table method,with 6 mice in each group.The SAMP8 blank group was randomly grabbed and bound without acupuncture every day,the SAMP8+Yuanluo Tongjing group was treated with needling Fenglong(ST40),Taibai(SP3),Taixi(KI3),Feiyang(BL58),and Baihui(DU20),and the SAMP8+Baihui group was only pricked at Baihui(DU20)once a day,with 30 interventions,Morris water maze and open field test were used to detect the cognitive and spatial learning and memory abilities of mice.HE staining was used to observe the pathological damage degree of neurons in hippocampal CA1 region.Apoptosis of hippocampal neurons was detected by TUNEL.The expressions of synaptophysin(SYN)and postsynaptic membrane protein-95(PSD-95)were detected by immunohistochemistry.The contents of Aβ_(40),p-tau in hippocampus were detected by ELISA.Results(1)Compared with the SAMP1 group,the escape latency and the escape path of the other three groups were prolonged,the time of staying in the second quadrant were shortened,the times of passing the original platform position and the times of entering the open field center were reduced,and the differences were statistically significant(P<0.05);Compared with SAMP8 blank group,the escape latency of mice in Yuanluo Tongjing group and Baihui group were shortened,the time of staying in the second quadrant were prolonged,the times of passing the original platform position and entering the central area of the open field were increased,and the improvement of mice in Yuanluo Tongjing group was better than that in B
