摘 要:目的 比较血清hsa-miR-139-3p和hsa-miR-147b在肺腺癌及健康对照组中的表达水平差异,探索血清hsa-miR-139-3p和hsa-miR-147b对患者的诊断价值。方法 收集齐齐哈尔附属第三医院2020年1月-2021年1月就诊的20例肺腺癌患者和20名健康对照者的血清进行RNA提取。应用实时荧光定量聚合酶链反应(qRT-PCR)方法,检测血清中hsa-miR-139-3p和hsa-miR-147b的表达量。利用受试者工作特征(ROC)曲线评价血清hsa-miR-139-3p和hsa-miR-147b的诊断效能。结果 肺腺癌患者血清hsa-miR-139-3p和hsa-miR-147b的表达水平较健康对照组显著升高,差异有统计学意义(P<0.05)。ROC曲线显示血清hsa-miR-139-3p和hsa-miR-147b分别检测以及联合检测均能够有效区分非小细胞肺癌患者和健康人群,曲线下面积(AUC)分别为0.79、0.7625和0.8125。结论 血清hsamiR-139-3p和hsa-miR-147b有希望作为肺腺癌非侵入性的分子诊断标志物,但尚需要进一步比较研究和扩大验证。Objective To compare the expression levels of serum hsa-miR-139-3p and hsamiR-147b in lung adenocarcinoma and healthy controls,and to explore the diagnostic value of serum hsa-miR-139-3p and hsa-miR-147b in patients.Methods The serum of 20 patients with lung adenocarcinoma and 20 healthy controls from January 2020 to January 2021 in the Third Affiliated Hospital of Qiqihar were collected for RNA extraction.The expression levels of hsamiR-139-3p and hsa-miR-147b in serum were detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR),and the diagnostic efficacy of hsa-miR-139-3p and hsamiR-147b in serum was evaluated by the ROC curve.Results The expression levels of serum hsa-miR-139-3p and hsa-miR-147b in patients with lung adenocarcinoma were significantly higher than those in healthy controls(P<0.05).ROC curve shows that serum hsa-miR-139-3p and hsa-miR-147b can effectively distinguish non-small cell lung cancer patients from healthy people,and the areas under the curve(AUC)are 0.79,0.7625 and 0.8125 respectively.Conclusion Serum hsa-miR-139-3p and hsa-miR-147b are promising as non-invasive molecular diagnostic markers for lung adenocarcinoma,but further comparative study and verification are needed.
