清热化瘀与清热益气化瘀中药巴布膏对PGE2在家兔急性软组织损伤模型中的影响

摘　　要：目的比较清热化瘀与清热益气化瘀中药巴布膏对急性软组织损伤家兔的前列腺素E2(prostaglandin E2,PGE2)的影响。方法由广东省医学实验动物中心提供24只SPF级雄性家兔,建立家兔急性软组织损伤动物模型,将其编号后,随机分为空白组、造模组、清热化瘀巴布膏组(清巴组)以及清热益气化瘀巴布膏组(益巴组),观测其皮下瘀血、肌肉肿胀、肌肉颜色和疼痛反应,检测24 h、72 h时各组受损肌肉组织及血清的PGE2含量变化,观察24 h、72 h时各组肌肉组织HE染色并予评分。结果造模24 h、48 h、72 h时观测损伤症状评分,在损伤评分中造模后0 h,各组与空白组比较差异均有统计学意义(P<0.05),各造模组之间差异无统计学意义(P>0.05),在造模及给药24 h后,各组与空白组差异仍有统计学意义(P<0.05),而各组间无明显差异,在造模48 h后与模型组相比,益巴组差异具有统计学意义(P<0.05);模型组HE染色形态学观察评分较空白组差异有统计学意义(P<0.05)。益巴组的评分较模型组的差异有统计学意义(P<0.05);肌肉PGE2含量中24 h清巴组与72 h清巴组差异有统计学意义(P<0.05)。24 h益巴组与72 h益巴组差异有统计学意义(P<0.05),造模24 h后,与空白组对比,模型组差异有统计学意义(P<0.05),治疗组差异无统计学意义。与模型组对比,两种治疗方法也差异无统计学意义。造模72 h后,与空白组对比模型组、治疗组无统计学差异。与模型组对比也差异无统计学意义。两种治疗方法对比无统计学意义。24 h清巴与72 h清巴、24 h益巴与72 h益巴无差异。结论相比清热化瘀巴布膏,清热益气化瘀巴布膏对家兔软组织受损部位的PGE2抑制效果更好,对急性软组织损伤的修复和消炎镇痛有明显作用。Objective To compare the effects of Babu Ointment(巴布膏)for clearing heat and removing blood stasis and Babu Ointment for clearing heat and invigorating Qi and removing blood stasis on prostaglandin E2(PGE2)in rabbits with acute soft tissue injury.Methods Twenty-four SPF male rabbits were provided by Guangdong Medical Experimental Animal Center to es⁃tablish the animal model of acute soft tissue injury.The rabbits were randomly divided into blank group,model group,Babu Oint⁃ment for clearing heat and removing blood stasis group(Qingba group)and Babu Ointment for clearing heat and invigorating Qi and removing blood stasis group(Yiba group).The subcutaneous blood stasis,muscle swelling,muscle color and pain reaction were observed.The changes of PGE2 content in damaged muscle tissue and serum of each group were detected at 24 h and 72 h,and the HE staining and score of muscle tissue were observed at 24 h and 72 h.Results The injury symptom score was observed at 24 h,48 h and 72 h after modeling.0 h after modeling,there was significant difference of each group and blank group(P<0.05),but there was no significant difference among the modeling groups(P>0.05).After 24 hours of modeling and administra⁃tion,there was still significant difference of each group and the blank group(P<0.05),but there was no significant difference a⁃mong the groups.Compared with the model group 48 hours after modeling,there was significant difference in Yiba group(P<0.05).The morphological observation score of HE staining in the model group was significantly higher than that in the blank group(P<0.05).The score of Yiba group was significantly higher than that of model group,and there was significant difference in muscle PGE2 content between 24 h and 72 h in Qingba group.There was significant difference between 24 h and 72 h in Yiba group(P<0.05).After 24 hours,compared with the blank group,there was significant difference in the model group(P<0.05),but there was no significant difference in the treatment groups.Compared with the mo
