血清外泌体let-7d-3p与miR-30d-5p在宫颈癌中的诊断价值

摘　　要：目的研究探索血清外泌体miRNA let-7d-3p与miR-30d-5p在宫颈癌患者中的表达,为宫颈癌早期诊断寻找新的肿瘤标志物。方法选取海南医学院第一附属医院2019年1月到2020年12月收治的宫颈癌患者与体检健康人群,采用qRT-PCR法检测血清外泌体let-7d-3p与miR-30d-5p的表达水平,ROC曲线分析目标基因在宫颈癌中的诊断价值,研究目标基因表达水平与宫颈癌患者临床特征的相关性。结果宫颈癌组血清let-7d-3p的表达水平是健康人群的1.95倍,miR-30d-5p表达水平宫颈癌组是健康人群的2.35倍。经检验差异均有统计学意义(P均<0.05)。二者联合诊断宫颈癌具有较好的诊断效能,曲线下面积为0.925。let-7d-3p与miR-30d-5p的表达水平与年龄和病理类型无关(P均>0.05),与肿瘤直径、淋巴结转移和FIGO分期呈正相关(P均<0.05)。结论血清外泌体let-7d-3p与miR-30d-5p联合对宫颈癌的早期诊断具有一定临床价值,其表达水平与肿瘤进展有关。Objective The study explores the expression of serum extract miRNA let-7d-3p and miR-30d-5p in cervical cancer patients,looking for new tumor markers for early diagnosis of cervical cancers.Methods Selected were the healthy individuals and cervical cancer patients admitted to the First Affiliated Hospital of the Hainan Medical University between January 2019 and December 2020.qRT-PCR was used to determine the amounts of let-7d-3p and miR-30d-5p expression in the serum exocrine.The diagnostic utility of the target gene in cervical cancer was examined using the ROC curve,and a correlation between the expression level of the target gene and the clinical characteristics of cervical cancer patients was investigated.Results The level of expression of the serum of the cervical cancer group let-7d-3p is 1.95 times that of the healthy population,and the level of miR-30d-5p expression is 2.35 times as of the average population.The test differences are statistically significant(P both<0.05).The two jointly diagnosed cervical cancers have better diagnostic effectiveness,accumulating 0.925 below the curve.The expression level of let-7d-3p and miR-30d-5p are irrelevant to age and type of pathology(P>0.05)and are correlated with tumor diameter,lymph nodes transfer and FIGO segmentation(P<0.05).Conclusion Serum lets-7d-3p,in combination with miR-30d-5p,has a specific clinical value for early diagnosis of cervical cancer,and its level of expression is associated with tumor progression.
