摘  要:目的:观察小核仁RNA宿主基因12(SNHG12)对裸鼠人肾透明细胞癌(ccRCC)移植瘤生长的影响。方法:选取福建省漳州市医院泌尿外科2021年12月至2022年10月经肾癌根治术患者的ccRCC及癌旁肾组织标本各11例,进行实时荧光定量反转录聚合酶链反应(RT-qPCR)检测SNHG12的表达;构建敲减SNHG12慢病毒,获得稳定表达敲减SNHG12慢病毒的786-0细胞株(si-SNHG12组)及稳定表达阴性慢病毒的786-0细胞株(NC组);选取对数生长期的si-SNHG12组和NC组的786-0细胞株建立裸鼠人ccRCC移植瘤模型,21 d后分离肿瘤组织,测量肿瘤的重量和体积,组间比较采用t检验。结果:ccRCC组织(5.925±4.380)中SNHG12的表达量显著高于癌旁肾组织(1.011±0.012),差异有统计学意义(t=3.721,P<0.01);获得稳定表达敲减SNHG12慢病毒及稳定表达阴性慢病毒的786-0细胞株:si-SNHG12组(0.296±0.007)中SNHG12的表达量显著低于NC组(1.000±0.036),差异有统计学意义(t=-33.290,P<0.01);si-SNHG12组(0.171±0.030)的移植瘤体重显著小于NC组(0.461±0.030),差异有统计学意义(t=-19.368,P<0.01)。si-SNHG12组(288.460±37.609)的移植瘤体积显著小于NC组(731.212±145.882),差异有统计学意义(t=-8.312,P<0.01)。结论:敲减SNHG12能够抑制裸鼠人肾透明细胞癌移植瘤的生长。Objective To observe the effect of small nucleolar RNA host gene 12(SNHG12)on the growth of human clear cell renal cell carcinoma(ccRCC)xenografts in nude mice.Methods A total of 11 ccRCC and 11 adjacent renal tissue samples from patients who underwent radical nephrectomy for renal cell carcinoma from December 2021 to October 2022 in the Department of Urology,Zhangzhou Hospital,Fujian Province were selected.The real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR)was used to detect the expression of SNHG12.The knockdown SNHG12 lentivirus was constructed to obtain 786-0 cell line stably expressing knockdown SNHG12 lentivirus(si-SNHG12 group)and 786-0 cell line stably expressing negative lentivirus(NC group).The 786-0 cell lines in the logarithmic growth phase si-SNHG12 group and NC group were selected to establish a human ccRCC xenograft model in nude mice,and tumor tissues were isolated 21 days later to measure the weight and volume of tumors.Results SNHG12 expression was significantly higher in ccRCC tissues(5.925±4.380)than in adjacent renal tissues(1.011)±0.012),and the difference was statistically significant(t=3.721,P<0.01).In the 786-0 cell line that obtained stable expression knockdown SNHG12 lentivirus and stable negative lentivirus,the expression level of SNHG12 in the si-SNHG12 group(0.296±0.007)was significantly lower than that in the NC group(1.000±0.036),and the difference was statistically significant(t=-33.290,P<0.01).The xenografts in the si-SNHG12 group(0.171±0.030)weighed significantly less than in the NC group(0.461±0.030),and the difference was statistically significant(t=-19.368,P<0.01).The volume of transplanted tumors in the si-SNHG12 group(288.460±37.609)was significantly smaller than that in the NC group(731.212±145.882),and the difference was statistically significant(t=-8.312,P<0.01).Conclusion Knockdown of SNHG12 can inhibit the growth of human renal clear cell carcinoma xenografts in nude mice.
