摘 要:目的:探讨微小核糖核酸34a(miR-34a)及沉默信息调节因子1 (SIRT1)在宫颈癌组织中的表达及其相关性。方法:收集住院患者标本,采用RT-qPCR法检测miR-34a在正常宫颈、宫颈癌组织中的表达;免疫组化法检测SIRT1在宫颈癌组织及正常宫颈组织中的表达,分析miR-34a及SIRT1表达与宫颈癌患者临床病理参数的关系,并作Spearman相关分析。结果:miR-34a在宫颈癌组织中的表达明显低于正常宫颈组织(P<0.05),且miR-34a在宫颈癌组织的表达与肿瘤分化程度显著相关(均P<0.05);免疫组化显示SIRT1在宫颈癌组织中的表达高于正常宫颈组织,其阳性表达率分别为84%与20%,差异具有统计学意义(P<0.05);Spearman分析显示,SIRT1与miR-34a在宫颈癌组织中的表达呈负相关(r=-0.327,P<0.05)。结论:miR-34a在宫颈癌的发生中起重要作用,这为宫颈癌临床治疗提供了一个潜在靶点。Objective:To investigate the expression of and association of microRNA-34a(miR-34a)and silent information regulator 1(SIRT1)in cervical cancer tissues.Methods:Samples of inpatients were collected,and the expression of miR-34a in normal cervix and cervical cancer tissues was detected by RT-qPCR.The expression of SIRT1 in cervical cancer tissues was detected by immunohistochemistry,and the relationship between the expression of miR-34a,SIRT1 and clinicopathologic parameters of patients with cervical cancer was analyzed.Spearman correlation analysis was performed.Results:The expression of miR-34a in cervical cancer tissues was significantly lower than that in normal cervix(P<0.05).The expression of miR-34a in cervical cancer tissues was significantly correlated with the degree of tumor differentiation(P<0.05).Immunohistochemistry showed that the expression of SIRT1 in cervical cancer tissues was higher than that in normal cervix(P<0.05),and the positive expression rates were 84%and 20%,respectively.Spearman analysis showed that SIRT1 was negatively correlated with miR-34a expression in cervical cancer tissues(r=-0.327,P<0.05).Conclusion:MiR-34a plays an important role in the development of cervical cancer,which provides a potential target for clinical treatment of cervical cancer.
