摘 要:目的:探索丙醇二酸对秀丽隐杆线虫体内脂肪的影响及调节机制。方法:在NGM培养基中加入不同浓度丙醇二酸饲养线虫,油红O和尼罗红染色法展示线虫体内脂肪颗粒大小、数量和相对脂肪含量,测定线虫身体摆动频率、吞咽频率、48 h存活率、身宽、体长等指标。QPCR检测fat-1至fat-7、sbp-1和nhr-49基因相对表达量,荧光显微镜观察FAT-6:GFP、FAT-7:GFP荧光强度的变化。结果:10、50、100μg/mL丙醇二酸处理对线虫脂滴大小无影响,脂肪颗粒数量降至对照组的0.69、0.53、0.54倍,相对脂肪含量降至0.87、0.81、0.80倍。此外,身体摆动频率、吞咽频率、48 h后存活率和体长无显著差异,身宽分别降至53.11、57.01、56.03μm。FAT-6:GFP、FAT-7:GFP荧光强度无明显变化,nhr-49、sbp-1、fat-1、fat-5、fat-6、fat-7基因转录水平无明显变化,fat-2、fat-3、fat-4基因转录水平分别降至0.69、0.77、0.24倍。结论:丙醇二酸可降低秀丽隐杆线虫体内脂肪,但不影响运动、吞咽等行为。降脂的调控不依赖于油酸转化,推测通过降低fat-2、fat-3、fat-4基因转录使得多不饱和脂肪酸(PUFAs)减少导致脂肪含量降低。Objective To explore hydroxymalonic acid on the influence of body fat and regulating mechanism in C.elegans.Method Nematodes were feed on the NGM culture medium with different concentration of hydroxymalonic acid,Oil red O and Nile red staining were used to show the size,number and relative fat content of fat particles in nematodes.At the same time,Body swinging frequency,swallowing frequency,48 h survival rate,body width and length were measured.The relative expressions of fat-1 to fat-7,sbp-1 and nhr-49 genes were detected by QPCR,and the changes of fluorescence intensity of FAT-6:GFP and FAT-7:GFP were observed by fluorescence microscope.Result Result showed that 10,50,and 100μg/mL hydroxymalonic acid treatment had no effect on lipid droplets size,but fat particles number dropped to 0.69,0.53,and 0.54 times,relatively fat decrease to 0.87,0.81,and 0.80 times.In addition,There was no significant difference in body swing frequency,swallowing frequency,survival rate and body length after 48 hours.Nevertheless body width respectively dropped to 53.11,57.01 and 56.03μm.The fluorescence intensity of FAT-6:GFP and FAT-7:GFP did not change significantly,and the transcriptional levels of nhr-49,sbp-1,fat-1,fat-5,fat-6 and fat-7 genes has no obvious change,but fat-2,fat-3,fat-4 gene transcription level dropped to 0.69,0.77,and 0.24 times respectively.Conclusion Hydroxymalonic acid reduced the body fat of Caenorhabditis elegans,but did not affect the behavior of exercise,swallowing and so on.The regulation of lipid lowering does not depend on oleic acid transformation.It is speculated that the decrease of polyunsaturated fatty acids(PUFAs)leads to the decrease of fat content by reducing the transcription of fat-2,fat-3 and fat-4 genes.
