摘 要:目的探索USP19调控结肠癌细胞侵袭转移的作用和机制。方法通过组织标本免疫组化染色和生物信息学方法分析验证USP19在结肠癌组织中的作用,通过Transwell小室检测USP19对结肠癌细胞侵袭、迁移能力的影响,通过基因敲低及过表达实验验证USP19相关调控的分子机制。结果25对结肠癌临床标本的免疫组化染色和蛋白印迹实验结果均显示USP19在结肠癌组织中显著高表达,通过TCGA数据库预后分析发现USP19高表达组的结肠癌患者10年总生存率显著低于USP19低表达组,提示USP19高表达与结肠癌不良预后密切相关。通过细胞实验发现USP19可促进结肠癌细胞侵袭转移,对其分子机制探索发现USP19可通过调控肿瘤细胞上皮-间充质转化(EMT)来增强结肠癌细胞侵袭能力和迁移能力。结论USP19在结肠癌侵袭转移相关调控中起关键作用,为结肠癌转移的具体调控机制提供了新的线索和思路。Objective To explore the mechanism of USP19regulating the invasion and metastasis of colon cancer cells.Methods Immunohistochemical staining and bioinformatics analysis were used to verify the role of USP19in colon cancer tissues.Transwell assay was used to detect the effect of USP19on invasion and migration of colon cancer cells.Knockdown plasmid transfection and Western blot were used to determine the molecular mechanism of USP19signaling.Results Immunohistochemical staining and Western blot results of 25pairs clinical colon cancer specimens showed that USP19was significantly overexpressed in cancer tissues,and TCGA database prognostic analysis showed that the 10-year overall survival rate of USP19 high-expressed group was significantly lower than that of USP19low-expressed group,suggesting that USP19high expression is closely related to the poor prognosis of colon cancer.We found that USP19could promote the invasion and metastasis of colon cancer cells in cellular phenotype experiments.The exploration of its molecular mechanism showed that USP19could increase the invasion and migration ability of colon cancer cells by regulating epithelial-mesenchymal transition(EMT)of tumor cells.Conclusion This study found a key role of USP19in the regulation of colon cancer invasion and metastasis,and provided new clues and ideas for the specific regulatory mechanism of colon cancer metastasis.
