摘 要:DNA嵌入剂通过非共价作用进入到DNA碱基对,可改变DNA的二级结构,影响DNA的性质.已有DNA嵌入剂多为有机分子,而具有丰富功能性的多金属氧簇DNA嵌入剂尚未报道.本文通过共价修饰将三种具有DNA嵌入能力的有机基团连接到Anderson型多金属氧簇上,并用紫外-可见吸收光谱、荧光光谱和圆二色光谱对其与DNA的作用强度进行评估.研究表明,在空间位阻和嵌入作用的共同影响下,含有苯并菲咯啉四酮的多金属氧簇杂化物与DNA具有最强的结合能力,结合常数达到10~4L mol^(-1).透射电子显微镜和原子力显微镜结果表明嵌入结合对多金属氧簇杂化物和DNA的形貌都有显著影响.本文的工作将DNA嵌入剂研究扩展到了多金属氧簇领域,为更多功能性DNA嵌入剂的开发提供了新思路.DNA intercalator can enter the middle of DNA base pairs through the non-covalent interaction,which changes the secondary structure of DNA and affects its properties.Most of the reported DNA intercalators are organic molecules,and polyoxometalates with rich properties have not been involved yet.In this article,three organic groups with DNA intercalation ability are linked to Anderson-type polyoxometalate through a covalent modification,and their interaction strengths with DNA are evaluated through UV-Vis spectra,fluorescence spectra,and circular dichroic spectra.Studies show that under the joint influence of steric hindrance and intercalation,polyoxometalate hybrid containing benzophenanthroline tetraketone has the strongest binding ability to DNA,and the binding constant reaches 10~4L mol~(-1).The results of transmission electron microscopy and atomic force microscopy show that intercalation binding has a significant impact on the morphology of polyoxometalate hybrid and DNA.This work extends the study of DNA intercalators to the field of polyoxometalates,and also provides a new idea for the development of DNA intercalators.
