摘 要:目的:探讨NOP56(Nucleolar Protein 56)在肝细胞癌(HCC)中的临床意义及潜在机制。方法:利用HCCDB及Oncomine数据库对多个队列的研究分析NOP56的表达;从癌症基因组图谱(TCGA)下载肝细胞癌数据集及临床数据,分析NOP56 mRNA表达水平与临床指标的相关性;运用Kaplan-Meier plotter在线分析NOP56对肝细胞癌患者预后的影响,并基于多因素Cox回归模型探索独立危险因素。利用GeneMANIA数据库构建以NOP56为核心的基因网络,并使用WebGestalt数据库分析这一网络预测NOP56的靶向药物;使用R软件的clusterProfiler包对NOP56正负相关基因进行基因富集分析(GSEA)。结果:相对于正常组织,NOP56 mRNA在肝细胞癌的表达上调;NOP56 mRNA的表达与肿瘤分级、肝癌家族史及肝细胞癌周围炎症相关;NOP56 mRNA高表达与预后差相关,并且NOP56 mRNA的表达是总体生存期的独立危险因素;NOP56互作网络富集于核糖体生物发生和rRNA代谢过程,并且放射菌素D可能作为NOP56的靶向药物;对NOP56正负相关基因进行富集分析得出NOP56可能影响核糖体及细胞周期。结论:NOP56是评估肝细胞癌患者的一种新型的预后指标,可能为潜在的治疗靶标。Objective:To investigate the clinical significance and potential mechanism of NOP56(Nucleolar Protein 56)in hepatocellular carcinoma.Methods:HCCDB and Oncomine databases were used to explore the expression of NOP56 through the study of multiple queues.The NOP56 mRNA expression and clinical data of hepatocellular carcinoma were downloaded from the cancer Genome Map(TCGA)to analyze the correlation between the expression level of NOP56 mRNA and clinical indicators.Kaplan-Meier Plotter was used to analyze the influence of NOP56 on the prognosis of patients with hepatocellular carcinoma online,and Cox proportional hazard regression model was used to analyze the risk factors affecting the prognosis of patients.Using GeneMANIA database to build NOP56 as the core gene network,and using WebGestalt database to analyze this network to predict NOP56 targeted drugs;the algorithm GSEA was applied to analyze NOP56 positive and negative related genes with R software clusterProfiler package.Results:The expression of NOP56 mRNA in hepatocellular carcinoma was up-regulated compared with normal tissue;NOP56 mRNA expression was associated with tumor grade,family history of hepatocellular carcinoma,as well as adjacent hepatic tissue inflammation;high expression of NOP56 mRNA is associated with poor prognosis,and NOP56 mRNA expression is an independent risk factor for overall survival;the NOP56 interaction network is enriched in ribosomal biogenesis and rRNA metabolism,and radiobacterin D may be a targeted drug for NOP56;enrichment analysis of NOP56 positive and negative related genes showed that NOP56 may affect ribosome and cellcycle.Conclusion:NOP56 may be a new prognostic indicator for patients with hepatocellular carcinoma,indicating that it may be a potential therapeutic target and may promote the development of hepatocellular carcinoma by influencing ribosome and cell cycle.
