摘 要:目的观察SLC5A8基因敲除后皮下种植结肠癌细胞株MC-38的负瘤模型小鼠肿瘤微环境中T淋巴细胞亚群占比变化,为结肠癌基因治疗提供实验依据。方法应用CRISPR/Cas9基因编辑技术构建SLC5A8基因敲除型C57BL/6小鼠,记为实验组。将普通C57BL/6小鼠记为对照组。将两组小鼠皮下种植结肠癌细胞株MC-38细胞悬液制备MC-38负瘤模型,制备成功后完整剥离肿瘤组织,采用流式细胞法测算各组小鼠结肠肿瘤微环境中T淋巴细胞亚群占比。结果实验组小鼠肿瘤微环境中CD3^(+)T淋巴细胞、CD4^(+)T淋巴细胞、CD8^(+)T淋巴细胞占比分别为27.730%±2.861%、50.923%±4.823%、10.205%±1.234%,对照组小鼠肿瘤微环境中CD3^(+)T淋巴细胞、CD4^(+)T淋巴细胞、CD8^(+)T淋巴细胞占比分别为40.790%±9.940%、58.355%±3.292%、22.950%±1.948%,两组相比,P均<0.05。结论与普通C57BL/6小鼠相比,SLC5A8基因敲除后皮下种植MC-38细胞的负瘤模型小鼠肿瘤微环境中CD3^(+)T淋巴细胞、CD4^(+)T淋巴细胞、CD8^(+)T淋巴细胞占比均下降。Objective To observe the changes in the proportion of T lymphocyte subsets in tumor microenvironment of tumor-bearing model mice with subcutaneous implantation of colon cancer cell line MC-38 after SLC5A8 gene knockout,and to provide experimental basis for colon cancer gene therapy.Methods SLC5A8 knockout C57BL/6 mice were constructed using CRISPR/Cas9 gene editing technology and were recorded as the experimental group.Normal C57BL/6 mice were recorded as the control group.The cell suspension of colon cancer cell line MC-38 was implanted subcutaneously into mice of the two groups to prepare the MC-38 tumor-bearing models,and the tumor tissues were completely stripped after successful preparation.The proportion of T lymphocyte subsets in the colon tumor microenvironment of mice in each group was measured by flow cytometry.Results The percentages of CD3^(+)T lymphocytes,CD4^(+)T lymphocytes,and CD8^(+)T lymphocytes in the tumor microenvironment of the experimental group were 27.730%±2.861%,50.923%±4.823%,and 10.205%±1.234%,respectively.The proportions of CD3^(+)T lymphocytes,CD4^(+)T lymphocytes,and CD8^(+)T lymphocytes in tumor microenvironment of mice in the control group were 40.790%±9.940%,58.355%±3.292%,and 22.950%±1.948%,respectively.There were significant differences between these two groups(all P<0.05).Conclusion Compared with the normal C57BL/6 mice,the proportions of CD3^(+)T lymphocytes,CD4^(+)T lymphocytes,and CD8^(+)T lymphocytes decreased in tumor microenvironment of tumor-bearing model mice implanted with MC-38 cells after SLC5A8 gene knockout.
