摘　　要：通过胃腺癌肺转移模型确定SPINT3与胃癌转移之间的相关性.结果表明, SPINT3 sgRNA在胃腺癌肺部转移中的表达均为阳性;SPINT3的表达与转化生长因子β1(TGFβ1)的分泌密切相关.检测SPINT3与肿瘤细胞的凋亡、增殖、迁移、体内转移能力和肿瘤相关通路的关联, SPINT3在MKN-45细胞中的过表达能够促进肿瘤细胞的凋亡,抑制增殖与迁移. SPINT3的抑制促进了MKN-45的体内转移能力, SPINT3的表达特异性的调节了TGFβ1通路.通过药物筛选确定了SPINT3的低表达能够被阿帕替尼和SB431542所逆转,这两种药物能够促进MKN-45 SPINT3 shRNA的凋亡,阻断体内转移.A correlation between SPINT3 and gastric cancer metastasis was determined via the lung me-tastasis model of gastric adenocarcinoma,the expression of SPINT3 sgRNA was positive in lung metas-tasis nodules and the expression of SPINT3 was closely related to the secretion of TGFβ1.The relation-ship was detected between SPINT3 and apoptosis,proliferation,migration,in vivo metastasis and tumor related pathways.The results showed that the overexpression of SPINT3 in MKN-45 cells could promote the apoptosis,inhibit proliferation and migration of tumor cells,while the inhibition of SPINT3 promoted the in vivo metastasis ability of MKN-45,and SPINT3 specifically regulated the TGFβ1 pathway.Through drug screening,it was determined that the low expression of SPINT3 could be reversed by Apa-tinib and SB431542,which could promote the apoptosis of MKN-45 SPINT3 shRNA and block metasta-sis invivo.
