摘 要:为探究宁夏野生岩羊(Pseudois nayaur)小反刍兽疫病毒(Peste des petits rumi-nants virus,PPRV)的分子遗传特征,根据Gen Bank公布的PPRV基因组序列设计并合成13对全基因组扩增引物,通过RT-PCR和DNA测序及拼接获取毒株(wild-bharal/China-NXYH/2021株)全基因组序列,借助DNASTAR和MEGA 7.0软件进行序列分析。结果显示:该毒株属于基因Ⅳ系,基因组全长为15954 nt;在系统进化上,与2013年以来中国流行株和2016—2017年蒙古流行株亲缘关系较近,共同组成一个小的进化分支;在全基因组一致性分析中,与国内新疆流行株goat/China-XJYL/2013一致性最高(99.4%),与国内其他野生动物PPRV株wild-bharal/China-Tibet/2008、Ibex/China-XJBZ/2015和Procapra-przewalskii/China-GS/2018的一致性分别为96.8%、98.8%和99.0%。在H和F蛋白氨基酸多态性分析中,发现该毒株H和F蛋白中均出现了独特的氨基酸突变,分别为H蛋白的R285Q、S421R和F蛋白的R/K/T3W、Q305R。结果表明,小反刍兽疫病毒已传入宁夏野生动物种群,为我国小反刍兽疫根除工作带来新的困难和挑战,需加强对宁夏野生动物PPRV感染情况的追踪和监测,以便及时从传染源和传播途径方面阻断PPRV的传播。In order to explore the molecular genetic characteristics of peste des petits ruminants virus(PPRV)isolated from wild bharal(Pseudois nayaur)in Ningxia,13 pairs of whole genome amplification primers were designed and synthesized ac⁃cording to the PPRV genome sequence published on GenBank.The whole genome sequence of the virus strain(wild-bharal/China-NXYH/2021 strain)was obtained through RT-PCR,DNA sequencing and splicing,and its sequence was analyzed with DNASTAR and MEGA 7.0 software.The results showed that the strain belonged to gene IV strain,with a total genome length of 15,954 nt.In terms of phylogenetic evolution,it was close to the Chinese epidemic strains since 2013 and the Mon⁃golian epidemic strains from 2016 to 2017,forming a small evolutionary branch.In the genome wide identity analysis,the highest identity(99.4%)was found with the domestic Xinjiang epidemic strain goat/China-XJYL/2013,and 96.8%,98.8%and 99.0%with other domestic wild animal PPRV strains wild-bharal/China-Tibet/2008,Ibex/China-XJBZ/2015 and Procapra-przewalskii/China-GS/2018,respectively.In the analysis of amino acid polymorphism of H and F proteins,unique amino acid mutations were found in both H and F proteins of the strain,namely R285Q and S421R of H protein and R/K/T3W and Q305R of F protein.The results showed that the peste des petits ruminants virus had been introduced into the wildlife population of Ningxia,bringing new difficulties and challenges to the eradication of peste des petits ruminants in China.The results suggest that it is necessary to strengthen the tracking and monitoring of PPRV infection of wild animals in Ningxia,so as to timely block the transmission of PPRV from the source of infection and transmission routes.
