摘 要:以1994年利用无菌水保藏的44株黑木耳菌种和同时期同种4℃继代保藏黑木耳菌种为试材,采用生长状态测定、ISSR(inter-simple sequence repeat)、酯酶同工酶的方法,研究无菌水保藏方法对黑木耳菌种性状的影响,以期对黑木耳菌种的无菌水保藏方法优劣的判断提供参考依据。结果表明:44株黑木耳在无菌水中常温保藏28年,有36株保持活性,复活率达到81.8%,菌种继代培养后菌丝恢复正常生长状态,并能够正常出耳;与4℃保藏菌种相比,同一菌株间酯酶同工酶和ISSR,总体具有一致性,个别菌株具有一定的差异,不同菌株反映不同。Forty-four strains of Auricularia heimuer preserved in sterile water in 1994 and the same strains of Auricularia heimuer preserved at 4℃C in the same period were used as experimental materials.The effects of sterile water preservation methods on the traits of Auricularia heimuer were studied by growth state determination,ISSR(inter-simple sequence repeat)and esterase isozyme,in order to provide reference for the judgement of the sterile water preservation methods of Auricularia heimuer.The results showed that 44 strains of Auricularia heimuer were preserved in sterile water at room temperature for 28 years,36 strains remained active,and the reactivation rate reached 81.8%.After subculture,the mycelium returned to normal growth state and could produce ears normally.Compared with the strains preserved at 4 C,the esterase isozyme and ISSR of the same strain were generally consistent,individual strains had certain differences,and different strains reflected different.
