一株寄生于大黄鱼的盾纤毛虫分子鉴定与系统进化分析

摘　　要：在大黄鱼稚鱼上发现一种可侵入患鱼体表、肌肉、腹腔和脑的盾纤毛虫,为了进一步明确该盾纤毛虫的分类地位,提取患鱼样品总DNA,对该虫株SSU rDNA和线粒体cox1部分序列进行PCR扩增、测序,与GenBank中纤毛虫的SSU rDNA和线粒体cox1序列进行同源性分析并构建系统发育树。结果显示,该盾纤毛虫874 bp的SSU rDNA部分序列与显赫针口虫(Porpostoma notata)同源性最高,相似性为98.97%。在基于SSU rDNA构建系统发育树中与显赫针口虫(P.notata)聚为一支,并独立于嗜污科(Philasteridae)的分支;获得该虫1 086 bp的cox1部分序列,与其SSU rDNA序列相比表现出更大的遗传变异度,在基于cox1部分序列构建系统发育树中,该虫株与嗜污科(Philasteridae)、尾丝虫科(Uronematidae)的分支也有一定距离。综上,该盾纤毛虫应是嗜污目(Philasterida)、针口虫属(Porpostoma)的显赫针口虫(P.notata)或其近缘种。This study was arm to further clarify the taxonomic status of a scuticociliate which can invade the surface,muscle,abdominal cavity and brain of juvenile large yellow croaker.Total DNA of sick fish sample was extracted.Then,partial sequences of SSU rDNA and the mitochondrial cox1 gene were amplified by PCR and sequenced.Similarity and the phylogenetic relationship between the sequences of the scuticociliate and the references in GenBank were analyzed by bioinformatics softwares.The results showed that the 874 bp partial SSU rDNA sequence had the highest homology with Porpostoma notata(98.97%similarity).In the phylogenetic tree based on SSU rDNA,it was clustered into a lineage with P.notata and independent from the branch of Philasteridae.Moreover,1086 bp partial cox1 sequence of the scuticociliate was amplified and sequenced.Compared with SSU rDNA sequences,cox1 sequences showed greater genetic variation.In phylogenetic tree based on cox1 partial sequences,it also separated from the branches of Philasteridae and Uronematidae.To sum up,it is suggested that the scuticociliate is P.neotata or related species of Porpostoma,Philasterida.
