摘  要:目的探讨DNA损伤激活的非编码RNA(NORAD)在口腔鳞状细胞癌组织,癌旁组织及口腔鳞癌细胞系中的表达及作用。方法回顾性选取2020年1月至2022年1月在河北医科大学第四医院行手术切除的口腔鳞状细胞癌组织标本88例,同时选取癌旁组织标本88例作为对照。采用PCR检测组织中NORAD mRNA相对表达量,分析不同临床病理特征口腔鳞状细胞癌组织NORAD mRNA相对表达量差异;同时选取口腔鳞癌细胞系SCC9、CAL27、SCC15及TSCCA细胞,以及正常黏膜HOK细胞,检测各细胞NORAD mRNA相对表达量差异。结果口腔鳞状细胞癌组织NORAD mRNA相对表达量为2.21±0.87,明显高于癌旁组织(1.03±0.34),差异有统计学意义(P<0.05)。TNM分期Ⅲ~Ⅳ期、低分化、有淋巴结转移口腔鳞状细胞癌组织NORAD mRNA相对表达量分别为2.79±0.69、2.62±0.65、2.68±0.68,明显高于TNM分期Ⅰ~Ⅱ期、中高分化、无淋巴结转移口腔鳞状细胞癌组织(1.98±0.67、2.03±0.70、1.93±0.62),差异均有统计学意义(P<0.05);复发和未复发患者NORAD mRNA相对表达量比较,差异无统计学意义(P>0.05)。CAL27细胞NORAD mRNA相对表达量为1.87±0.41,明显高于SCC15、SCC9、TSCCA、HOK细胞(1.40±0.44、1.39±0.46、1.06±0.34和0.51±0.18),差异均有统计学意义(P<0.05)。结论NORAD在口腔鳞状细胞癌组织表达上调,与TNM分期、分化程度及淋巴结转移有关,NORAD在CAL27细胞中表达最高。Objective To investigate the expression and role of DNA damage activated non coding RNA(NORAD)in oral squamous cell carcinoma tissues,adjacent tissues and oral squamous cell carcinoma cell lines.Methods Eighty-eight oral squamous cell carcinoma tissue samples were selected from January 2020 to January 2022,and 88 specimens of tissues adjacent to cancer were selected as controls,PCR was used to detect the relative expression of NORAD mRNA in tissues,the relative expression of NORAD mRNA in oral squamous cell carcinoma tissues with different clinicopathological characteristics was analyzed.And oral squamous cell carcinoma cell lines SCC9,CAL27,SCC15 and TSCCA,as well as normal mucosal HOK cells were selected to detect the difference in the relative expression of NORAD mRNA in each cell.Results The relative expression of NORAD mRNA in oral squamous cell carcinoma was 2.21±0.87,which was significantly higher than that in the adjacent tissues(1.03±0.34),the difference was statistically significant(P<0.05).The relative expression of NORAD mRNA in TNM stageⅢ~Ⅳ,poorly differentiated and lymph node metastatic oral squamous cell carcinoma tissues was 2.79±0.69,2.62±0.65 and 2.68±0.68,respectively,which were significantly higher than those in TNM stageⅠ-Ⅱ,moderately well differentiated and non lymph node metastatic oral squamous cell carcinoma tissues(1.98±0.67,2.03±0.70,1.93±0.62),the differences were statistically significant(P<0.05).There was no significantly significant difference in the relative expression of NORAD mRNA between relapsed and non relapsed patients(P>0.05).The relative expression level of NORAD mRNA in CAL27 cells was 1.87±0.41,which was significantly higher than those in SCC15,SCC9,TSCCA,and HOK cells(1.40±0.44,1.39±0.46,1.06±0.34,and 0.51±0.18),the differences were statistically significant(P<0.05).Conclusion NORAD is up-regulated in oral squamous cell carcinoma,which is related to TNM stage,differentiation and lymph node metastasis.NORAD is the highest expression in CAL27 cells.
