摘 要:[目的]研究外源一氧化氮(Nitric oxide,NO)对马铃薯块茎创伤木栓化过程中内源激素含量的影响,为筛选影响马铃薯块茎愈合的内源激素,并深入研究马铃薯块茎创伤愈合中内源激素反应机制提供科学依据。[方法]本试验以‘滇薯47’为试验材料,洗净并去除马铃薯块茎的表皮后,用打孔器和刀片将马铃薯块茎处理为直径约15 mm、厚度约5 mm的小圆片,重量为1 g左右。实验设置3个处理,CK处理:超纯水;硝普钠处理(Sodium nitroprusside,SNP):0.5 mmol/L SNP(NO供体);C-PTIO处理:1.3 mmol/L C-PTIO(NO清除剂),并放置于温度(23±1)℃,空气相对湿度85%,光照为0的培养箱中进行创伤愈合反应,测定创伤后不同时间块茎中与马铃薯块茎创伤愈合相关的内源激素含量。[结果]外源NO处理(SNP处理)引起创伤后马铃薯块茎中内源激素含量变化。外源NO处理能促进创伤块茎中脱落酸(Abscisic acid,ABA)、乙烯前体1-氨基环丙烷-1-羧酸(1-aminocyclopropane-1-carboxylic acid,ACC)、水杨酸(Salicylic acid,SA)和生长素(Auxin,IAA)等激素含量升高,而C-PTIO处理则表现出抑制作用;外源NO处理在一定程度上促使内源茉莉素(Jasmonate,JA)类的含量降低,而C-PTIO处理可促进内源JA类含量增加,表明NO可能是JA的负调控因子;外源NO可促进细胞分裂素类物质中反式玉米素(Trans zeatin,tZ)、顺式玉米素核苷(Cis zeatin riboside,czR)、异戊烯基腺苷(Isopentenyl adenosine,iPA)含量的增加,对顺式玉米素(Cis zeatin,cZ)、反式玉米素核苷(Trans zeatin riboside,tzR)、异戊烯基腺嘌呤(Isopentenyl adenine,iP)的促进作用不明显,但C-PTIO处理对cZ、tzR、czR、iP和iPA含量的上升在后期有抑制作用。[结论]外源NO处理可以调控创伤马铃薯块茎中内源激素含量变化,加快创面愈合速度。外源NO处理可以提高马铃薯创伤块茎中ABA、ACC、SA、IAA等内源激素的含量,抑制JA含量增长,从而加快创伤块茎的愈合速度。[Objective]The effects of exogenous Nitric oxide(NO)on endohormone contents in suberification of potato tuber wound were studied to provide a scientific basis for screening out endohormone in tuber healing and further study on the mechanism of endohormone in tu-ber wound healing of potato.[Method]In this experiment,potato cultivar D47'was used as the material.,After washing and removing the pericarp of the potato tuber,the potato tuber was cut into smallround pieces with a diameter of about 15 mm,thickness of 5 mm and weight of 1 g by using a hole puncher and a blade.Three treatments were set up in the experiment,CK treatment:Ultra-pure water;Sodium nitro-prusside(SNP)treatment:0.5 mmol/L SNP(NO donor);C-PTIO treatment:1.3 mmol/L C-PTIO(NO scavenger),and they were placed in a light temperature incubator[dark,(23±1)℃,85%humidity]for wound healingreaction.After 0,27,46,54,72hours,the endogenous hormones content were determined during wound healing to study the mechanism of endogenous hormones involved in potato tuber healing.[Result]Exogenous NO treatment(SNP treatment)caused changes of endogenous hormone content in potato tuber after trau-ma.Exogenous NO treatment could promote Abscisic acid(ABA),ethylene precursor 1-aminocyclopropane-1-carboxylic acid(ACC),Sali-cylic acid(SA)in wound tubers.Auxin(IAA)and other hormones increased,whereas C-PTIO showed inhibitory effect.Exogenous NO treatment reduced the content of endogenous Jasmonate(JA)to a certain extent,while C-PTIO treatment promoted the increase of endoge-nous JA content,suggesting that NO might be a negative regulator of JA.Exogenous NO could promote the increase of contents of Trans zeat-in(tZ),Cis zeatin riboside(czR)and Isopentenyl adenosine(iPA)in cytokinins.The promotion effect on Cis zeatin(cZ),Trans zeatin riboside(tzR),Isopentenyl adenine(iP)was not obvious.However,C-PTIO treatment inhibited the content increase of cZ,tzR,czR,iP and iPA in the later period.[Conclusion]Exogenous NO treatment could regulate the content of endogenous hormones in the
