摘 要:为提高野金柴老叶中根皮苷的浸出率,对其复合酶预处理工艺进行了研究。在构成复合酶的纤维素酶、果胶酶和淀粉酶单因素试验基础上,将纤维素酶量(A)、果胶酶量(B)以及淀粉酶量(C)的使用量作为影响因子,以经过酶处理后的干叶用沸水冲泡后其根皮苷的释放量(R1)为响应值,通过Design Expert 10软件的响应面Box-Behnken试验设计,对上述3因子及其交互作用进行了研究。结果表明:A,B的交互作用对R1影响均达到了极显著水平,C的交互作用不显著;野金柴老叶干叶复合酶预处理最佳工艺为纤维素酶的用量为干叶重量的0.2%、果胶酶的用量为1.0%、α-淀粉酶的用量为0.97%,经此处理后的野金柴干叶用沸水冲泡后其根皮苷释放量可达18423 mg/kg,与最高预测值18933 mg/kg比较接近,表明该模型能较好地预测实际处理情况。In order to improve the extraction rate of phloside from Lithocarpus polystachyus Rehd,the pretreatment technology of complex enzyme was studied.Based on single factor test of cellulase,pectinase and amylase,the amount of cellulase(A),pectinase(B)and amylase(C)were taken as the influencing factors.The extraction amount of phlorhizin(R1)of the enzyme-treated dry leaves after boiling water was taken as the response value.Above three factors and their interactions were studied through the response surface Box-Behnken test design by Design Expert 10 software.The results showed the interaction of A and B had significant effects on R1,while the C did not.The optimal treatment technology of composite enzymes was determined as follows:the dosage of cellulase,pectinase andα-amylase was at 0.2%,1.0%and 0.97%of the dry leaves weight,respectively.The extraction amount of phlorhizin from the treated dry leaves was reaching 18423 mg/kg.It was close to the highest predicted value of 18933 mg/kg,which indicating the model could predict the actual treatment.
