摘  要:目的观察Foxp1在两种模型小鼠脑内反应性星形胶质细胞中的表达差异。方法利用脂多糖(LPS)、大脑中动脉栓塞(MCAO)构建LPS模型和MCAO模型小鼠,诱导两种模型小鼠脑内产生反应性星形胶质细胞;通过免疫荧光染色方法,使用胶质纤维酸性蛋白(GFAP)、离子钙结合适配器分子1(Iba1)、神经元特异性核蛋白(NeuN)分别标记模型小鼠脑内阳性的星形胶质细胞、小胶质细胞及神经元,并结合苏木精-伊红染色观察细胞形态,鉴定模型是否构建成功;采用免疫荧光双标方法检测Foxp1与GFAP或Iba1在两种模型小鼠大脑皮层(CTX)、纹状体(STR)、海马(HIP)3个脑区中的共标情况。结果两种模型小鼠构建成功,并且成功诱导小鼠脑内产生反应性星形胶质细胞;Foxp1和GFAP双阳性细胞数在LPS模型小鼠的CTX、STR、HIP3个脑区均明显增加(P<0.05),而在MCAO模型小鼠的CTX和STR区域中均明显减少(P<0.05),在HIP区域中无明显变化;在LPS模型及MCAO模型小鼠的STR区域中均未检测到Foxp1和Iba1双阳性细胞。结论Foxp1在LPS模型小鼠脑内的反应性星形胶质细胞中表达上升,而在MCAO模型中表达降低。Objective To observe the expression difference of Foxp1 in reactive astrocytes in the brain of two mouse models.Methods Reactive astrocytes were induced by lipopolysaccharide(LPS)and cerebral ischemia(middle cerebral artery occlusion,MCAO)models.To verify that the two models were constructed successfully,immunofluorescence staining was employed to label glial fibrillary acidic protein(GFAP)-positive astrocytes,ionized calcium binding adapter molecule 1(Iba1)-positive microglia and neuron-specific nuclear protein(NeuN)-positive neurons,and hematoxylin and eosin(HE)staining was used to observe the morphological changes of cells.Co-labeling of Foxp1 with astrocyte marker GFAP or microglia marker Iba1 in cortex(CTX),striatum(STR)or hippocampus(HIP)of model mice were detected by immunofluorescence double staining.Results The LPS and MCAO mouse models were successfully constructed,and reactive astrocytes were successfully induced in the brain of two models.The number of Foxp1 and GFAP double-positive cells was significantly increased in all three brain regions of CTX,STR and HIP in LPS-induced model mice(P<0.05),while it was significantly reduced in the CTX and STR of the MCAO models(P<0.05),with no significant change in the HIP.Foxp1 and Iba1 double-positive cells were not detected in the STR of the two models.Conclusion The expression of Foxp1 is increased in LPS-induced reactive astrocytes and decreased in MCAO induced reactive astrocytes.
