金雀异黄酮对口腔癌体外放射治疗的有效性研究

摘  要:目的观察不同剂量的X射线照射后,金雀异黄酮干预治疗对人永生化皮肤表皮角质形成细胞(HaCaT)、人正常肺上皮细胞(Beas-2B)、人舌癌细胞(Tca-8113)、人口腔鳞状细胞(HCS-4)的抑制率和凋亡情况,探讨金雀异黄酮联合X射线照射对口腔癌治疗的有效性。方法取人HaCaT、Beas-2B、Tca-8113、HCS-4细胞进行常规体外培养,将细胞分为4组:对照组(未加金雀异黄酮且不进行X射线照射)、金雀异黄酮组、X射线组和金雀异黄酮+X射线组。其中,金雀异黄酮组将金雀异黄酮(浓度分别为1.25、2.50、5.00、10.00、20.00μmol/L)加入4种细胞中处理12 h;X射线组用X射线(剂量分别为1、3、7 Gry)对4种细胞照射24 h;金雀异黄酮+X射线组根据X射线剂量不同,得到4种细胞的金雀异黄酮最佳处理值,用流式细胞仪和噻唑蓝法测定4种细胞的凋亡水平和细胞抑制率,并进行分析。结果4种细胞加入不同浓度的金雀异黄酮后,HaCaT和Tca-8113两种细胞抑制率提高,差异有统计学意义(P<0.001),且与加入的金雀异黄酮浓度呈正相关,在20.00μmol//L浓度下细胞抑制率达到峰值。4种细胞进行不同能量值X射线照射后,仅有HSC-4细胞抑制率高于其他3种细胞,差异有统计学意义(P<0.001),且与X射线剂量呈正相关,当剂量为7 Gry时达到最高。仅有HaCaT细胞经金雀异黄酮和射线照射后细胞凋亡水平提高,差异有统计学意义(P<0.001)。金雀异黄酮组HaCaT、Tca-8113细胞的凋亡水平高于其他两种细胞(P<0.001),但经射线照射后,HaCaT、Beas-2B、HSC-4细胞的凋亡水平有所提高(P<0.001)。与其他3组比较,金雀异黄酮+X射线组对HSC-4细胞的抑制作用更强,且细胞凋亡水平更高,差异有统计学意义(P<0.001)。结论金雀异黄酮与X射线联合作用能有效抑制HCS-4和Tca-8113细胞的增殖,促进癌细胞凋亡,二者联用对口腔癌治疗有效。Objective To explore the efficacy of genistein combined with X-ray in the treatment of oral cancer by observing the effect of genistein treatment on the cell inhibition rate and apoptosis of human immortalized skin epidermal keratinocytes(HaCaT),human normal lung epithelial cells(Beas-2B),human tongue cancer cells(Tca-8113)and human oral squamous cells(HCS-4)under different doses of X-ray irradiation.Methods HaCaT,Beas-2B,Tca-8113 and HCS-4 cells were cultured in vitro and divided into 4 groups:control group(without genistein treatment and X-ray irradiation),Gen group,X-ray group and Gen+X-ray group.In the Gen group,the four kinds of cells were treated by genistein(concentrations 1.25、2.50、5.00、10.00 and 20.00μmol/L,respectively)for 12 h.In the X-ray group,the four kinds of cells were irradiated for 24 h by X-ray(doses 1,3 and 7 Gry,respectively).In the Gen+X-ray group,the optimal treatment values of genistein in different concentrations for oral cancer cells were obtained according to the different X-ray doses.The apoptotic state and cell inhibition rate of the four kinds of cells were measured by flow cytometry and MTT assay,and analyzed.Results The apoptosis levels of HaCaT and Tca-8113 cells were increased after the treatment of different concentrations of genistein(P<0.001),and were positively correlated with the concentration of genistein,and peaked at the concentration of 20.00 umol/L.After X-ray irradiation of the four kinds of cells grouped by different energy values,only HSC-4 cells showed a higher inhibition rate than the other three kinds of cells(P<0.001),which was positively correlated with the X-ray dosage,and peaked when the dosage was 7 Gry.Only HaCaT cells displayed increased apoptosis levels after treatment with genistein and radiation(P<0.001).The apoptosis level of HaCaT and Tca-8113 cells in the Gen group was higher than that of the other two kinds of cells(P<0.001),but after X-ray treatment,the iapoptosis level of HaCaT,Beas-2B and HSC-4 cells was increased(P<0.001).Compared with the
