摘　　要：目的:探讨PRL-3在AML中的表达及其临床预后价值。方法:使用GEPIA2分析PRL-3在AML患者中的表达变化;利用UALCAN进一步进行亚组分析;采用OncoLnc分析PRL-3表达水平与AML患者总生存期的关系并验证,评估预后;通过STRING分析PRL-3相关基因及其蛋白互作网络,并采用Metascape进行GO和KEGG富集分析。结果:PRL-3在AML患者中的表达较正常人低(P<0.05)。各亚型中,PRL-3在MO和M6中的表达相对较高,在M3和M5中的表达较低;且在61~80岁患者中的表达明显高于41~60岁(P<0.05)。PRL-3高表达患者的总体生存期较低表达明显缩短(P<0.05)。此外,在AML中与PRL-3相互作用蛋白且低表达的主要是BCAR1、MAPK1、RHOC。PRL-3及其相关基因参与的生物学功能和通路涉及肿瘤细胞黏附、迁移、运动等方面。结论:PRL-3可能通过参与髓外浸润影响AML患者预后,可成为AML诊断和预后的潜在靶点。Objective:To investigate the expression of PRL-3 in AML and its clinical prognostic value.Methods:GEPIA2 was used to analyze the expression changes of PRL-3 in AML patients.UALCAN was used for further subgroup analysis.OncoLnc was used to analyze the relationship between PRL-3 expression level and overall survival of AML patients,and verify to evaluate the prognosis.PRL-3-related genes and their protein interaction networks were analyzed by STRING,GO and KEGG enrichment analysis was performed using Metascape.Results:The expression of PRL-3 in AML patients was lower than that in normal subjects(P<0.05).In each subtype,the expression ofPRL-3 was relatively high in MO and M6,and the expression in M3 and M5 was low.The expression in patients aged 61-80 was significantly higher than that in patients aged 41-60(P<0.05).The overall survival time of patients with high PRL-3 expression was significantly shortened(P<0.05).In addition,BCAR1,MAPK1,and RHOC were mainly low-expressed proteins that interact with PRL-3 in AML.The biological functions and pathways involved in PRL-3 and its related genes involve tumor cell adhesion,migration,and movement.Conclusion:PRL-3 may ffect the prognosis of AML patients by participating in extramedullary infiltration,and may become a potential target for the diagnosis and prognosis of AML.
