摘　　要：目的:研究DEP结构域蛋白1B(DEPDC1B)对肝癌细胞增殖、迁移和上皮间质转化的作用以及在临床中的相关性。方法:收集39例肝癌肿瘤组织及其癌旁组织,qRT-PCR检测DEPDC1B表达水平并分析与临床病理特征的相关性,通过Kaplan-Meier数据库分析DEPDC1B表达水平与临床预后的相关性。采用qRT-PCR检测L-02正常肝上皮细胞和HepG2肝癌细胞DEPDC1B表达水平,Western Blot检测DEPDC1B、E-cadherin、N-cadherin蛋白表达水平。将si-DEPDC1B和si-NC转染至HepG2细胞,采用qRT-PCR检测DEPDC1B表达水平,Western Blot检测DEPDC1B、E-cadherin、N-cadherin蛋白表达水平,采用CCK8和集落形成实验检测细胞增殖水平,Transwell和细胞划痕实验检测细胞迁移水平。结果:与癌旁组织相比,肿瘤组织中DEPDC1B表达水平升高,DEPDC1B高表达与肿瘤分期和淋巴结转移相关,与治疗后的不良预后相关。与L-02细胞相比,HepG2细胞DEPDC1B表达升高,N-cadherin蛋白表达升高,E-cadherin蛋白表达降低。与si-NC组比较,si-DEPDC1B组DEPDC1B表达降低,细胞增殖、迁移水平降低,N-cadherin蛋白水平降低,E-cadherin蛋白表达升高。结论:DEPDC1B可以促进肝癌细胞增殖、迁移和上皮间质转化,临床上DEPDC1B高表达与肿瘤晚期、淋巴结发生转移以及不良预后相关。Objective:To investigate the effect of DEP domain protein 1B(DEPDC1B)on proliferation,migration and epitheliaL-mesenchymal transition(EMT)of hepatocellular carcinoma cells and its clinical relevance.Methods:39 cases of HCC tissues and adjacent tissues were collected.The expression level of DEPDC1B was detected by qRT-PCR,and the correlation between DEPDC1B expression level and clinicopathological features was analyzed.The correlation between DEPDC1B expression level and clinical prognosis was analyzed by Kaplan-Meier database.The expression levels of DEPDC1B in L-02 normal liver epithelial cells and HepG2 hepatoma cells were detected by QRT-PCR,and the protein expression levels of DEPDC1B.E-cadherin and N-cadherin were detected by WesternBlot.Si-DEPDC1B and si-NC were transfected into HepG2 cells,and the expression of DEPDC1B was detected by qRT-PCR.The protein expression levels of DEPDC1B,E-cadherin,and N-cadherin were detected by WesternBlot.CCK8 and colony formation assay were used to detect cell proliferation,and Transwell and cell scratch assay were used to detect cell migration.Results:The expression level of DEPDC1B was increased in tumor tissues compared with adjacent tissues.High expression of DEPDC1B was associated with tumor stage and lymph node metastasis,and was associated with poor prognosis.Compared with L-02 cells,the expression of DEPDC1B,N-cadherin and E-cadherin in HepG2 cells was increased,while the expression of E-cadherin was decreased.Compared with si-NC group,the expression of DEPDC1B,cell proliferation,migration and N-cadherin protein were decreased in si-DEPDC1B group,while the expression of E-cadherin protein was increased.Conclusion:DEPDC1B can promote the proliferation,migration and epitheliaL-mesenchymal transition of hepatocellular carcinoma cells.High expression of DEPDC1B is associated with advanced tumor stage,lymph node metastasis and poor prognosis.
