摘 要:为研究白背清风藤(Sabia discolor Dunn)的化学成分及其对降血糖活性的体外抑制作用。该文采用硅胶柱层析、凝胶柱层析和半制备液相色谱等方法对白背清风藤进行分离纯化,运用^(1)H NMR、^(13)C NMR和MS等波谱技术对化合物进行结构鉴定;并对所有化合物的α-葡萄糖苷酶的体外抑制作用进行测定。结果表明:(1)从白背清风藤干燥枝叶中分离得到16个五环三萜类化合物,分别鉴定为:3α-羟基-齐墩果酸甲酯(1)、二羟基蒲公英烷(2)、(20S)-3-oxo-20-hydroxytaraxastane(3)、白桦脂醇(4)、20-taraxastene-3α,28-diol(5)、sabiadiscolor A(6)、1-oxo-erythrodiol(7)、3-epi-betulin(8)、蒲公英烷-3β,20α-二醇(9)、1β,2α,3β-trihydroxylup-20(29)-ene(10)、paracaseolin A(11)、sabiadiscolor C(12)、3β-hydroxyolean-12-en-28-oic acid(13)、乌苏酸(14)、olean-2,12-dien-1-one(15)和juglangenin A(16)。其中化合物1、8、10、11、12、13和15为首次从该植物中分离得到。(2)药理活性结果表明,化合物5、8和11均对α-葡萄糖苷酶具有显著的抑制活性,IC_(50)值分别为(16.73±1.23)、(20.06±1.39)和(17.87±3.49)μmol/L,抑制活性与阳性药阿卡波糖相当(IC_(50)=26.23±1.37μmol/L)。该研究结果丰富了白背清风藤降血糖的活性物质基础,为其进一步开发利用提供了科学依据。In order to explore the chemical constituents of Sabia discolor Dunnand their inhibitory effects on hypoglycemic activity in vitro.The chemical constituents of S.discolor Dunn were isolated and purified by silica gel column chromatography,Sephadex LH-20 column chromatography andsemi-preparative HPLC,and their structures were identified by means of various spectroscopic techniques such as ^(1)H NMR,^(13)C NMR and MS.An in vitro screening model was employed to test theα-glucosidase activity of obtained compounds.The results showed as follows:(1)Sixteen compounds were isolated from S.discolor Dunn and identified as3α-hydroxy-oleanolic acid methyl ester(1),dihydroxy-dandelane(2),(20S)-3-oxo-20-hydroxytaraxastane(3),betulinol(4),20-taraxastene-3α,28-diol(5),sabiadiscolor A(6),1-oxo-erythrodiol(7),3-epi-betulin(8),dandelane-3β,20α-diol(9),1β,2α,3β-trihydroxyl-20(29)-ene(10),paracaseolin A(11),sabiadiscolor C(12),3β-hydroxyolean-12-en-28-oic acid(13),ursolic acid(14),olean-2,12-dien-1-one(15)and juglangenin A(16).Compounds 1,8,10,11,12,13 and 15 were isolated from this plant for the first time.(2)The results of activity test showed that 20-taraxastene-3α,28-diol(5),3-epi-betulin(8)and paracaseolin A(11)had good inhibitory effects onα-glucosidase in vitro,with IC_(50) values of(16.73±1.23),(20.06±1.39),and(17.87±3.49)μmol/L,respectively,whose activitieswere equivalent to the positive drug acarbose(IC_(50)=26.23±1.37μmol/L).This study enrich the material basis of hypoglycemic activity of S.discolor Dunn,providing scientific basis for its further development and utilization.
