摘　　要：磷酸二酯酶4(PDE4)是神经退行性疾病的一个可观的药物靶点。尽管大量临床前研究表明PDE4抑制剂对中枢神经系统(CNS)疾病存在有益作用,但目前尚无CNS适应症的药物。在这篇综述中,我们讨论了PDE4抑制剂用于治疗阿尔茨海默症以及帕金森等其他神经退行性疾病的效果,包括常见的PDE4抑制剂、新型研发的PDE4抑制剂、PDE4抑制剂与其他药物的多靶点用药以及PDE4抑制剂和其他PDE家族药物的联合用药等等。Phosphodiesterase 4(PDE4)is a promising drug target for neurodegenerative diseases.Although numerous preclinical studies have shown beneficial effects of PDE4 inhibitors on central nervous system(CNS)diseases,there are currently no drugs for CNS indications.In this review,we discuss the efficacy of PDE4 inhibitors in the treatment of Alzheimer's disease and other neurodegenerative diseases such as Parkinson's disease,including common PDE4 inhibitors,newly developed PDE4 inhibitors,multi-target use of PDE4 inhibitors with other drugs,and combination of PDE4 inhibitors with other PDE family drugs.
