摘  要:目的 研究血红素加氧酶-1(HO-1)及其抑制剂锌原卟啉Ⅸ在光动力治疗(PDT)胰腺癌中的作用,从而为光动力治疗胰腺癌的敏感性提供依据。方法 CCK-8法检测PDT及锌原卟啉Ⅸ(ZnPPⅨ)对胰腺癌细胞株PANC-1增殖能力的影响;利用Western blot和Real-time PCR技术分别检测胰腺癌细胞中HO-1蛋白及其mRNA的表达情况;构建胰腺癌裸鼠皮下种植瘤模型,进一步观察PDT及ZnPPⅨ对胰腺癌成瘤能力的影响。结果 PDT联合ZnPPⅨ干预胰腺癌细胞能明显抑制胰腺癌细胞的增殖率(P <0.05);PDT胰腺癌细胞中HO-1 mRNA水平和蛋白表达水平均增加,而PDT联合ZnPPⅨ干预后的胰腺癌细胞HO-1表达明显降低(P <0.05);动物实验结果显示,PDT联合ZnPPⅨ干预可抑制胰腺癌皮下种植瘤的生长,肿瘤体积和质量明显降低。结论 血红素加氧酶-1抑制剂ZnPPⅨ可增强胰腺癌光动力治疗的敏感性,可能成为胰腺癌光动力治疗的潜在增敏剂。Objective To investigate the role of heme oxygenase-1(HO-1) and its inhibitor zinc protoporphyrin Ⅸ in photodynamic therapy(PDT) of pancreatic cancer,so as to provide evidence for the sensitivity of photodynamic therapy of pancreatic cancer.Methods CCK-8 method was used to detect the effects of PDT and zinc protoporphyrin Ⅸ(ZnPPⅨ) on the proliferation of pancreatic cancer cell line PANC-1;Western blot and real time PCR were used to detect the expression of HO-1 protein and its mRNA in pancreatic cancer cells;The pancreatic cancer xenocrafts in nude mice were used to further observe the effect of PDT and ZnPPⅨ on the tumorigenicity of pancreatic cancer.Results PDT combined with ZnPPⅨ intervention can significantly inhibit the growth rate of pancreatic cancer cells(P<0.05);Both HO-1 mRNA level and protein expression level were found to be increased in PDT pancreatic cancer cells.However,HO-1 expression of pancreatic cancer cells after PDT combined with ZnPPⅨ intervention was significantly decreased(P<0.05).The results of animal experiments showed that PDT combined with ZnPPⅨ intervention inhibited the growth of the pancreatic cancer xenocrafts,and the tumor volume and mass were significantly reduced.Conclusion ZnPPⅨ,an inhibitor of heme oxygenase-1,can enhance the sensitivity of photodynamic therapy of pancreatic cancer,and may become a potential sensitizer for photodynamic therapy of pancreatic cancer.
