摘  要:为了对樱花中嫩红素成分进行准确定性、定量分析以及高效提取工艺研究。本研究以中国南方主栽樱花品种‘关山’为试验材料,利用HPLC-MS/MS法对其花瓣干品内的嫩红素成分进行定性、定量分析,并利用乙醇提取法、低温干燥法获得樱花冻干粉。结果表明,在HPLC-MS/MS分析中,在正离子模式下共检测出27种黄酮类物质(总黄酮相对浓度为44.85μg/mL),含量较高的有槲皮素-3-O-丙二酰基葡萄糖苷、O-丙二酰氨基己糖苷黄酮base+4O、槲皮素-3-β-D-葡萄糖甙等;在负离子模式下共检测出34种黄酮类物质(总黄酮相对浓度为1170.72μg/m L),含量较高的有芹菜素-7-(2"-乳酸葡糖酯)、猫眼草酚-C 6,4’-二葡糖苷、槲皮素-3-β-D-葡萄糖苷、6-羟基木犀草素7-(6"-丙二酰葡萄糖苷)等。樱花黄酮类物质成分测定在负离子模式下测定更为准确。此外,樱花嫩红素最佳提取工艺为:首先加8倍体积的70%乙醇没过样品浸泡1 h,再加入20倍体积70%乙醇进行渗漉提取,去除滤渣后低温真空浓缩,回收至无乙醇味,最后取浓缩液冷冻干燥获得樱花冻干粉,且冻干粉中黄酮类含量高达65.35%。研究结果为中国樱花资源的开发与应用提供一定的理论基础。In order to carry out accurate qualitative and quantitative analysis and high-efficiency extraction technology research on the pink magic components in cherry blossoms(Cerasus serrulata var.lannesiana).The main cherry variety C.serrulata var.lannesiana‘Kanzan’was used as the experimental material in southern China in this study.The pink magic components were qualitatively and quantitatively analyzed by HPLC-MS in its dried petals,and the freeze-dried powder of cherry blossoms was obtained by ethanol extraction and low-temperature drying.The results showed that the 27 flavonoids were detected in positive ion mode(the relative concentration of total flavonoids was 44.85μg/mL)in HPLC-MS/MS analysis,with higher content of quercetin 3-O-malonylglucoside,Flavone base+40,O-MalonylHex and quercetin-3-β-D-glucoside,etc..A total of 34 flavonoids were detected in the negative ion mode(the relative concentration of total flavonoids was 1170.72μg/mL),with higher contents of apigenin 7-(2"-glucosyllactate),chrysosplenol C 6,4'-diglucoside and quercetin-3-β-D-glucoside,etc..6-Hydroxyluteolin7-(6"-malonylglucoside)The determination of flavonoids was more accurate in the negative ion mode.In addition,the best extraction process of cherry blossom pink magic was as follows:first,add 8 times the volume of 70% ethanol to soak the sample for an hour,then add 20 times the volume of 70% ethanol for percolation extraction,remove the filter residue,and then concentrate in low temperature and vacuum,recover to no ethanol flavor,and finally take the concentrated solution for freeze-drying to obtain cherry blossom freeze-dried powder,and the content of flavonoids was as high as 65.35% in the freeze-dried powder.The research results provide a theoretical basis for the development and application of cherry blossom resources in China.
