摘  要:为研究植物甾醇酯(phytosterol ester,PE)抗炎及改善皮肤屏障的作用机制,通过十二烷基硫酸钠(sodium lauryl sulfate,SLS)作用于体外重组三维表皮模型(EpiKutis®)构建皮肤损伤模型,检测植物甾醇酯对组织活力、组织形态、炎症因子白介素1α (interleukin-1α,IL-1α)、肿瘤坏死因子α(tumor necrosis factor α,TNFα)、炎症介质前列腺素2(prostaglandin E2,PGE2)及屏障相关的丝聚蛋白(filaggrin,FLG)的影响。结果表明植物甾醇酯对SLS诱导的皮肤损伤模型的组织活力及组织形态无明显作用,对PGE2、IL-1α表达无显著抑制作用,但当其质量分数高于1%时能够显著抑制TNFα表达,并显著促进了FLG的表达。因此,植物甾醇酯具备一定抗炎及改善皮肤屏障的功效,但是PGE2和IL-1α不是植物甾醇酯抗炎的靶点。The aim of the study was to explore the mechanism on improving anti-inflammation and skin barrier of phytosterol ester,based on a EpiKutis®3D skin model which was specially damaged with sodium lauryl sulfate,by detecting the tissue viability,histological morphology,expression of interleukin 1α(IL-1α),tumor necrosis factorα(TNFα),inflammatory mediator prostaglandin E2(PGE2)and filaggrin(FLG).The results showed that phytosterol esters had neither significant positive effects on tissue viability and histological morphology based on SLS-indused skin model,nor inhibitions of PGE2 and IL-1αexpresions.However,it showed a significant inhibition of TNFαexpression and also a significant promotion of FLG expression,when its mass concentration was higher than 1%.Therefore,phytosterol ester has an anti-inflammatory and skin barrier improvement effects,and PGE2 and IL-1αare not anti-inflammatory targets of phytosterol esters.
