老年结直肠癌患者组织AIM2、hnRNP AB与血清CEA及病理参数的相关性

摘　　要：目的探讨老年结直肠癌患者组织黑色素瘤缺乏因子2(AIM2)、核不均一核糖核蛋白AB(hnRNP AB)和血清癌胚抗原(CEA)及病理参数的相关性。方法采集2017年6月至2020年6月收治的78例老年结直肠癌患者的癌组织及癌旁正常组织标本。采用免疫组化SP法检测癌组织及癌旁正常组织标本中的AIM2、hnRNP AB阳性表达情况,采用电化学发光法检测患者术前血清CEA水平。收集患者的临床病理参数,分析结直肠癌组织组织中AIM2、hnRNP AB表达及术前血清CEA与临床病理参数的关系。采用Kendall相关分析结直肠癌组织中AIM2、hnRNP AB表达与术前血清CEA的相关性。结果免疫组化显示,AIM2主要分布于肿瘤组织细胞质内,hnRNP AB主要分布于细胞核内;78例肿瘤组织中AIM2阳性29例(37.18%),明显低于癌旁组织的63例(80.77%),hnRNP AB阳性55例(70.51%),明显高于癌旁组织的17例(21.79%),术前CEA水平阳性34例(43.59%),明显高于癌旁组织的5例(6.41%),差异均有统计学意义(P<0.05)。结直肠癌组织中AIM2、hnRNP AB表达及术前血清CEA水平均与性别、年龄、肿瘤直径、肿瘤部位、分化程度、大体分型、远处转移等无关(P>0.05),而与浸润程度、TNM分期、淋巴结转移等有关(P<0.05)。Kendall相关分析显示,结直肠癌组织中AIM2表达与术前血清CEA水平呈负相关(r=-0.211,P<0.01),而结直肠癌组织中hnRNP AB表达与术前血清CEA水平呈正相关(r=0.291,P<0.01)。结论老年结直肠癌患者组织AIM2、hnRNP AB表达与血清CEA水平密切相关,浸润程度越深,TNM分期越晚,发生淋巴结转移的结直肠癌组织中AIM2表达降低、hnRNP AB表达升高,AIM2、hnRNP AB可能成为结直肠癌靶向治疗的新靶点。Objective To investigate the correlation between melanoma deficiency factor 2(AIM2),heterogeneous nuclear ribonucleoprotein AB(hnRNP AB),serum carcinoembryonic antigen(CEA)and pathological parameters in elderly patients with colorectal cancer.Methods Tissue samples from 78 elderly colorectal cancer patients admitted between June 2017 and June 2020 were collected,including cancer tissues and adjacent normal tissues.Immunohistochemical SP method was used to detect the positive expression of AIM2 and hnRNP AB in cancer tissues and adjacent normal tissues.The preoperative serum CEA levels of the patients were measured using electrochemiluminescence.Clinical pathological parameters of the patients were collected,and the relationship between AIM2,hnRNP AB expression in colorectal cancer tissues,preoperative serum CEA levels,and clinical pathological parameters was analyzed.Kendall correlation analysis was used to assess the correlation between AIM2,hnRNP AB expression in colorectal cancer tissues,and preoperative serum CEA levels.Results Immunohistochemistry showed that AIM2 was mainly distributed in the cytoplasm of tumor cells,while hnRNP AB was mainly distributed in the cell nucleus.Among the 78 tumor tissues,29 cases(37.18%)showed positive AIM2 expression,significantly lower than the adjacent tissues with 63 cases(80.77%).On the other hand,55 cases(70.51%)showed positive hnRNP AB expression,significantly higher than the adjacent tissues with 17 cases(21.79%).The preoperative CEA level was positive in 34 cases(43.59%),significantly higher than the adjacent tissues with 5 cases(6.41%).These differences were all statistically significant(P<0.05).AIM2,hnRNP AB expression in colorectal cancer tissues,and preoperative serum CEA levels were not associated with gender,age,tumor diameter,tumor location,differentiation degree,macroscopic type,distant metastasis(P>0.05),but were associated with infiltration depth,TNM stage,lymph node metastasis(P<0.05).Kendall correlation analysis showed that AIM2 expression in colorectal can
