摘 要:目的:探究利用电子舌技术对草乌进行麻味评价及其双酯型生物碱定量预测的可行性。方法:利用Half-tongue口尝方法评价不同质量浓度草乌提取液的麻味程度,确定麻味阈值,以阈值浓度的草乌提取液作参比样,用欧氏距离(D_(E))作参比样与各样品间电子舌区分的差别度,以判别电子舌检测中各样品的麻味程度,并利用高效液相色谱法(HPLC)测定草乌中双酯型生物碱含量,构建用D_(E)值预测双酯型生物碱含量的数据模型并验证。结果:基于口尝和电子舌检测结果,D_(E)值能够很好地反映各草乌提取液间的麻味差异,且D_(E)值和双酯型生物碱含量之间可用线性数据模型表示,其线性回归方程为:Y=7.222X+365.46(r=0.9949),该模型预测结果良好。结论:电子舌可以评价不同质量浓度草乌提取液的麻味程度,初步实现电子舌对草乌中双酯型生物碱的定量分析,为后期利用电子舌代替人工感官评价提供了理论和实践基础。Objective:To explore the feasibility of using electronic tongue technology to evaluate the Aconiti Kusnezoffii Radix numb-taste and quantitative prediction of numb-taste substances in Aconiti Kusnezoffii Radix.Methods:The half-tongue tasting method was used to evaluate the numb-taste degree of the extracts of Aconiti Kusnezoffii Radix with different mass concentrations,and the numb-taste threshold was determined.The threshold concentration of Aconiti Kusnezoffii Radix was used as the reference sample,and euclidean distance(D_(E))was used as the difference degree of electronic tongue discrimination between the reference sample and each sample.To distinguish the numb-taste degree of each sample in electronic tongue test.HPLC was used to determine the content of numb-taste substances in Aconiti Kusnezoffii Radix and a data model for predicting the content of numb-taste substances with D_(E)value was established and verified.Results:Based on the results of oral taste and electronic tongue detection,D_(E)value could well reflect the difference of numb-taste among the extracts of Aconiti Kusnezoffii Radix,and the relationship between D_(E)value and numb-taste substance content could be expressed by a linear data model.The linear regression equation was Y=7.222X+365.46(r=0.9949),and the prediction result of this model was good.Conclusion:Electronic tongue can evaluate the numb-taste degree of different mass concentrations of aconite extract,and preliminarily realize the quantitative analysis of diester alkaloids in aconite extract by electronic tongue,which provides a theoretical and practical basis for using electronic tongue instead of artificial sensory evaluation in the later period.
