摘  要:目的:探讨参芪复方对糖尿病KKAy小鼠味觉结构与功能的影响及其作用机制。方法:30只KKAy小鼠联合高脂高糖喂养建立糖尿病模型,随机分为模型组、二甲双胍组、参芪复方组,每组10只,每日分别予0.9%氯化钠溶液、二甲双胍、参芪复方灌胃;另设10只C57BL/6J小鼠作为正常组,每日予0.9%氯化钠溶液灌胃,连续8周。HE染色观察各组小鼠舌组织病理形态学改变,光镜下测量计算味蕾横截面积,实时荧光定量PCR检测T1R2、T1R3、SGLT-1 mRNA表达。结果:与正常组比较,模型组体质量显著增加(P<0.01);舌菌状乳头、味孔、味蕾数量减少,丝状乳头表层坏死脱落增加,真皮层内毛细血管充血出血,味蕾横截面积显著下降(P<0.05);T1R2、T1R3 mRNA表达呈上升趋势,SGLT-1 mRNA表达显著上调(P<0.01)。与模型组比较,各用药组体质量显著下降(P<0.01),舌组织病理损伤显著改善,参芪复方组味蕾横截面积显著增加(P<0.01);参芪复方组T1R2、T1R3 mRNA表达显著升高(P<0.01),且作用优于二甲双胍组(P<0.05);二甲双胍组SGLT-1 mRNA表达下调显著(P<0.05)。结论:糖尿病存在味觉感受系统的受损及甜味感知与传导能力的下降,参芪复方能提高味觉感知能力,改善味觉结构性损害,调节口腔甜味受体T1R2、T1R3表达,并下调SGLT-1表达,抑制葡萄糖转运吸收。Objective:To explore the effect and mechanism of Shenqi Compound(SQC)on the structure and function of taste in diabetic KKAy mice.Methods:Thirty KKAy mice were fed with high-fat and high-glucose to establish diabetes model,and were randomly divided into the diabetes group,the Glucophage group,and the SQC group,with 10 mice in each group.They were given normal saline,glucophage,and SQC intragastric administration respectively daily.10 C57BL/6J mice were set as the normal group,and they were given normal saline intragastric administration every day for 8 consecutive weeks.HE staining was used to observe the pathological changes of tongue tissue of the mice in each group,and the cross-sectional area of taste buds was measured and calculated under microscope.The mRNA expressions of T1R2,T1R3 and SGLT-1 were detected by real-time quantitative PCR.Results:Compared with the normal group,body weight of the diabetes group was increased significantly(P<0.01);the number of tongue fungus-like papillae,taste pores and taste buds decreased,and the surface necrosis and exfoliation of the filiform papillae increased,the capillaries were hyperemia and hemorrhaged in the corium layer,and the cross-sectional area of taste buds decreased(P<0.05);T1R2 and T1R3 mRNA expressions showed an upward trend,and SGLT-1 mRNA expression was significantly increased(P<0.01).Compared with the diabetes group,body weight of all drug groups was significantly decreased(P<0.01),the pathological changes of tongue tissue were significantly improved,and the cross-sectional area of taste buds in the SQC group was significantly increased(P<0.01);the mRNA expressions of T1R2 and T1R3 in all drug groups showed an upward trend,and the SQC group was significantly increased(P<0.01),and the effect was better than that in the Glucophage group(P<0.05);the mRNA expression of SGLT-1 showed a downward trend,and the Glucophage group decreased significantly(P<0.05).Conclusion:In diabetic mice,the taste perception system was impaired and the ability of sweet taste percep
