摘 要:目的探究miR-202-3p与血管内皮生长因子(VEGF)信号通路的调控关系及其影响肺腺癌细胞生长的分子机制。方法采用实时荧光定量PCR(qRT-PCR)检测细胞中miR-202-3p的表达水平。通过Western blot实验检测肺腺癌细胞中VEGF信号通路相关蛋白的磷酸化水平及蛋白水平。利用噻唑蓝(MTT)实验、细胞划痕实验及Transwell实验分别检测细胞的增殖、迁移与侵袭能力。结果miR-202-3p在肺腺癌细胞中显著下调。下调miR-202-3p提高了VEGF信号通路相关蛋白磷酸化水平,但不改变通路相关蛋白的表达,而加入通路抑制剂后通路相关蛋白磷酸化水平有所恢复。下调miR-202-3p可促进肺腺癌细胞的增殖、迁移与侵袭,加入VEGF信号通路抑制剂后,肺腺癌细胞的增殖、迁移与侵袭能力显著降低。结论miR-202-3p可调控VEGF信号通路抑制肺腺癌细胞的发生发展,miR-202-3p很可能是治疗肺腺癌的新靶点。Objective To explore the regulatory relationship between miR-202-3p and vascular endothelial growth factor(VEGF)signaling pathway and its molecular mechanism affecting the growth of lung adenocarcinoma cells.Methods The expression level of miR-202-3p in cells was detected by real-time fluorescence quantitative PCR(qRT-PCR).The phosphorylation level and protein level of VEGF signaling pathway related proteins in lung adenocarcinoma cells were detected by Western blot.The proliferation,migration and invasion of cells were detected by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT)assay,cell scratch assay and Transwell assay,respectively.Results miR-202-3p was significantly down-regulated in lung adenocarcinoma cells.Down-regulation of miR-202-3p increased the phosphorylation level of VEGF signaling pathway-related proteins,but did not change the expression of pathway-related proteins.The phosphorylation level of pathway-related proteins was restored after the pathway inhibitor was added.Down-regulating miR-202-3p could promotes the proliferation,migration and invasion of lung adenocarcinoma cells.After adding VEGF signaling pathway inhibitors,the proliferation,migration and invasion capabilities of lung adenocarcinoma cells were significantly reduced.Conclusion miR-202-3p could regulate the VEGF signaling pathway and inhibit the development of lung adenocarcinoma cells,miR-202-3p is likely to be a new target for the treatment of lung adenocarcinoma.
