摘  要:目的探讨中心体相关蛋白激酶2(NEK2)和细丝蛋白A(FLNa)的表达与胃癌患者临床特征及预后之间的关系。方法以2017年10月至2022年12月烟台市烟台山医院收治的108例胃癌患者组织标本作为研究对象, 采用免疫组织化学方法检测上述胃癌组织和癌旁组织中NEK2和FLNa表达水平, 结合临床资料, 应用χ^(2)检验进行分析。结果胃癌组织中NEK2表达率为62.96%(68/108), 明显高于癌旁组织中NEK2表达率[11.11%(12/108)], 差异有统计学意义(χ^(2)=62.259, P<0.01)。胃癌组织中FLNa表达率为30.56%(33/108), 明显低于癌旁组织中FLNa表达率[80.56%(87/108)], 差异有统计学意义(χ^(2)=54.675, P<0.01)。NEK2表达水平与胃癌患者组织学分化程度、TNM分期(TNM stage, tumor node metastasis stage)、淋巴结转移、肿瘤大小明显相关(χ^(2)=4.723、6.632、4.246、7.021, P<0.05), NEK2表达水平与胃癌患者年龄、性别无相关(χ^(2)=0.001、0.027, P>0.05)。FLNa表达水平与胃癌患者TNM分期、淋巴结转移、肿瘤大小明显相关(χ^(2)=5.102、6.279、5.645, P<0.05), FLNa表达水平与胃癌患者组织学分化程度、年龄、性别无相关(χ^(2)=0.004、0.023、0.329, P>0.05)。结论 NEK2在胃癌组织中高表达、FLNa在胃癌组织中低表达, NEK2和FLNa的表达水平与胃癌的形成发展密切相关。Objective To investigate the relationship between the expression of NIMA-related kinase 2(NEK2)and Filamin A(FLNa)in gastric cancer,and the clinical characteristics and prognosis of gastric cancer patients.Methods The tissue specimens of 108 patients with gastric cancer admited to Yantaishan Hospital from October 2017 to December 2022 were selected as the research objects.The ex-pression levels of NEK2 and FLNa in the above gastric cancer tissues and corresponding adjacent tissues were detected by immunohistochemistry.Combined with clinical data,Chi-square test was used for correla-tion analysis.Results The expression rate of NEK2 in gastric cancer tissues was 62.96%(68/108),which was significantly higher than that in adjacent tissues[11.11%(12/108),χ^(2)=62.259,P<0.01].The expression rate of FLNa in gastric cancer tissues was 30.56%(33/108),which was significantly lower than that in adjacent tissues[80.56%(87/108),χ^(2)=54.675,P<0.01].The expression level of NEK2 was significantly correlated with histological differentiation,tumor node metastasis(TNM)stage,lymph node metastasis and tumor size(respectively,χ^(2)=4.723,6.632,4.246,7.021,P<0.05),but not signif icantly with age and gender of gastric cancer patients(respectively,χ^(2)=0.001,0.027,P>0.05).The ex-pression level of FLNa was significantly correlated with TNM stage,lymph node metastasis and tumor size of gastric cancer patients(respectively,χ^(2)=5.102,6.279,5.645,P<0.05),but not significantly with histo logical differentiation,age and gender(respectively,χ^(2)=0.004,0.023,0.329,P>0.05).Conclusion NEK2 is highly expressed and FLNa is low expressed in gastric cancer tissues.The expression levels of NEK2 and FLNa are closely related to the formation and development of gastric cancer.NEK2 and FLNa can be used as important indicators for the prognosis of patients with gastric cancer.
