摘  要:目的 探讨β-榄香烯调节Notch1/Hes1信号通路对宫颈癌细胞增殖、凋亡和放射敏感性的作用机制。方法 将人宫颈癌SiHa细胞分为对照组(control)、β-榄香烯低、中、高剂量组(β-榄香烯-L:10μg/mL,M:25μg/mL,H:50μg/mL)组。采用CCK-8法检测宫颈癌细胞SiHa细胞增殖活性;细胞克隆形成实验SiHa细胞评估放射敏感性;Hoechst33258染色法检测SiHa细胞凋亡;划痕实验和Transwell实验检测SiHa细胞的迁移侵袭能力;Western blot检测凋亡蛋白、上皮间质转化蛋白、迁移侵袭蛋白和Notch1/Hes1信号通路蛋白表达。结果 与control组比较,β-榄香烯各处理组的SiHa细胞增殖活性、克隆形成率、迁移侵袭能力、蛋白Vimentin、N-cadherin、MMP-2、MMP-9、Notch1、Hes1表达显著下降(P<0.05),SiHa细胞的放射敏感性显著增强、Caspase-3、Caspase-9、E-cadherin表达显著提高(P<0.05)。结论 β-榄香烯可以下调Notch1/Hes1信号通路蛋白表达,从而抑制宫颈癌SiHa细胞增殖、迁移、侵袭,促进细胞凋亡,增强细胞放射敏感性。Objective To investigate the mechanism ofβ-elemene regulating Notch1/Hes1 signal pathway on prolif-eration,apoptosis and radiosensitivity of cervical cancer cells.Methods Human cervical cancer SiHa cells were divided into control group,low-dose,middle dose and high-doseβ-elemene groups(elemen-L:10μg/mL,M:25μg/mL,H:50μg/mL)group.The proliferation activity of cervical cancer cell SiHa was detected by CCK-8 method.The radiosensitivity of SiHa cells was evaluated by cell cloning assay.Hoechst 33258 staining was used to detect SiHa cell apoptosis.Transwell and scratch healing assays were performed to detect the cell migration and invasion.The expression of apoptosis protein,epithelial mesenchymal transformation protein,migration invasion protein and Notch1/Hes1 signal pathway protein were detected by Western blot.Results Compared with the control group,the proliferation activity,colony formation rate,migration and inva-sion ability of SiHa cells,the expression of Vimentin,N-cadherin,MMP-2,MMP-9,Notch1 and Hes1 proteins in theβ-elemene treated groups were greatly decreased(P<0.05).The radiosensitivity of SiHa cells was greatly enhanced,and the expression of Caspase-3,Caspase-9,E-cadherin was greatly increased(P<0.05).Conclusionβ-elemene can down regulate the expression of Notch1/Hes1 signal pathway protein,thereby inhibiting the proliferation,migration and invasion of cervical cancer SiHa cells,promoting cell apoptosis,and enhancing cell radiosensitivity.
