胶红酵母的分离鉴定与山苍子油的抑菌研究

摘　　要：从腐烂变质的花生中分离纯化出一株菌株E2,通过形态学、生理生化试验及26SrDNA测序进行鉴定.结果显示该菌株的26SrDNA测序序列与胶红酵母(Rhodotorula mucilaginosa)的同源性达100%,其形态特征及生理生化特性与胶红酵母最相似.通过测定抑菌圈直径、抑菌率和最低抑菌浓度(MIC)定性定量研究山苍子油对胶红酵母的抑菌活性,从离子渗漏试验和还原糖利用率探讨了山苍子油对胶红酵母细胞膜通透性的影响.试验结果表明,一定浓度的山苍子油能显著抑制胶红酵母的生长,最小抑菌体积分数(MIC)为1.200μL·mL^(-1),试验还表明山苍子油破坏了胶红酵母细胞膜结构,导致胞内离子流失,还原糖利用率显著降低,干扰其对营养物质的利用.A strain E2 was isolated and purified from decayed peanut.Morphological,physiological and biochemical tests,and 26SrDNA sequencing were used to classify and identify the strain.The results showed that the 26SrDNA sequencing sequence of this strain had 100%homology with Rhodotorula mucilaginosa,and its morphological,physiological and biochemical characteristics were most similar to Rhodotorula mucilaginosa.The antimicrobial activity of litsea cubeba oil against Rhodotorula mucilaginosa,was qualitatively and quantitatively studied by measuring the diameter of the bacteriostatic ring,the bacteriostatic rate,and the minimum inhibitory concentration(MIC).The effects of litsea cubeba oil on the permeability of Rhodotorula mucilaginosa cell membrane were discussed from the perspective of ion leakage test and reducing sugar utilization rate.The results showed that a certain concentration of litsea cubeba oil could significantly inhibit the growth of Rhodotorula mucilaginosa,with a minimum inhibitory volume fraction(MIC)of 1.200μL·mL-1,the test also showed that Litsea cubeba oil damaged the membrane structure of Rhodotorula mucilaginosa,resulting in the loss of intracellular ions,significantly reducing the utilization of reducing sugar,and interfering with its utilization of nutrients.
