健脾清化方对高脂饮食诱导肥胖小鼠胰岛细胞分化的影响

摘  要:目的分析健脾清化方对高脂饮食诱导肥胖小鼠胰岛细胞分化的影响。方法选用6周龄雄性C57BL/6小鼠适应性喂养1周后,随机分为正常组及高脂组。造模成功后,高脂组随机分为模型组及健脾清化方组,干预6周后检测各组小鼠体质量、空腹血糖(fasting bloodglucose,FBG)、空腹胰岛素(fasting insulin,FINS)、糖耐量、脂代谢指标,及苏木精-伊红染色观察小鼠胰腺组织形态和结构,并通过免疫荧光共染观察小鼠胰岛α细胞、β细胞及祖细胞标记物神经源素(neurogenin,Ngn)3的数量及分布。结果第5周,高脂组体质量较正常组显著增加(P<0.01),健脾清化方组体质量较模型组降低(P<0.05)。在注射葡萄糖30 min后,模型组较正常组血糖显著升高(P<0.01)。在注射葡萄糖30、60、90、120 min后,健脾清化方组血糖水平较模型组均显著降低(P<0.01),曲线下面积显著降低(P<0.01)。健脾清化方组FINS水平明显降低(P<0.01)。健脾清化方组较模型组血清胆固醇、三酰甘油水平降低(P<0.01)。荧光共染显示,健脾清化方组胰岛β细胞、α细胞明显增多,β细胞去分化标记-祖细胞标记物Ngn3明显减少。结论健脾清化方“从脾论治”,能有效降低高脂饮食诱导肥胖小鼠血糖、血脂及胰岛素水平,可能与其逆转胰岛β细胞去分化作用有关。Objective To analyze the effect of Jianpi Qinghua decoction on islet cell differentiation in obese mice induced by high fat diet.Methods Six-week-old male C57BL/6 mice were randomly divided into normal group and high fat diet group after adaptive feeding for 1 week.After successful modeling,the high-fat group was randomly divided into model group and Jianpi Qinghua decoction group.After 6 weeks of intervention,body mass,fasting blood glucose(FBG),fasting insulin(FINS),glucose tolerance and lipid metabolism indexes of mice in each group were measured.Hematoxylineosin staining was used to observe the morphology and structure of pancreatic tissue.Immunofluorescence co-staining was used to observe the quantity and distribution ofαcells,βcells and progenitor cell marker neurogenin(Ngn)3.Results At the 5th week,the body weight of the high fat group was significantly increased compared with the normal group(P<0.01),and the body weight of the Jianpi Qinghua decoction group was decreased compared with the model group(P<0.05).After 30 min of glucose injection,blood glucose of the model group was significantly higher than that of the normal group(P<0.01).After 30,60,90 and 120 min of glucose injection,the blood glucose level and area under the curve of Jianpi Qinghua decoction group were significantly decreased compared with model group(P<0.01).FINS levels in Jianpi Qinghua decoction group were significantly decreased(P<0.01).The levels of serum cholesterol and triacylglycerol in Jianpi Qinghua decoction group were lower than those in model group(P<0.01).Fluorescence co-staining showed thatβ-cell andα-cell in Jianpi Qinghua decoction group increased significantly,and Ngn3,a marker ofβ-cell dedifferentiation and progenitor cell,was significantly decreased.Conclusion Jianpi Qinghua decoction can effectively reduce the levels of blood glucose,blood lipid and insulin in obese mice induced by high fat diet,which may be related to reversing the dedifferentiation of pancreaticβcells.
