大米免疫活性肽凝胶色谱分离的研究

摘  要:以小鼠巨噬细胞增殖指数SI值(增殖指数:stimulating idex,SI)为考察指标,优化凝胶色谱分离纯化大米免疫活性肽的试验条件。结果表明,凝胶过滤色谱的试验条件为:洗脱剂蒸馏水;上样体积4 mL;上样浓度30 mg/mL;洗脱速度15 mL/h。在该条件下分离得到的4个洗脱组分的SI值均高于未经凝胶色谱分离的001×7-pH 7组分(SI值1.319,125μg/mL),其中F4组分的SI值最大,为1.518(125μg/mL),与001×7-pH 7组分相比,F4组分对小鼠巨噬细胞增殖具有极显著的促进作用(P<0.01)。Macrophage proliferation index SI was used as the index to research the conditions of gel isolation and purification for rice immuno-active peptide(RIAP).The results showed the conditions for gel filtration chromatography were as follows:eluent was distilled water,loading volume was 4 mL,sample concentration was 30 mg/mL,elution speed was 15 mL/h.It was found that the four elution components separated by Sephadex G-10 had higher activity compared with 001×7-pH 7 component(1.319,125μg/mL)which was separated without gel filtration chromatography.SI value of F4 component was 1.518(125μg/mL)which was the largest among all the components.Compared with the 001×7-pH 7 component,F4 had a very significant(P<0.01)promotion for the proliferation of mouse macrophage.
