摘 要:目的探究周期性牵张力对人牙周膜成纤维细胞(human periodontal ligament fibroblasts,hPDLFs)中骨形态发生蛋白-2(bone morphogenetic protein-2,BMP-2)表达的影响及对人牙周膜成纤维细胞(human periodontal ligament fibroblasts,hPDLFs)成骨分化的作用。方法体外培养hPDLFs,分为3组(空白对照组、6 h组、12 h组),分别不加力、加力6 h和12 h。使用免疫荧光实验检测3组细胞骨架蛋白表达情况;使用Western Blot实验检测3组BMP-2表达情况;碱性磷酸酶染色试剂盒及茜素红染色实验分别用于检测3组碱性磷酸酶(alkaline phosphatase,ALP)活性及骨钙结节形成情况。结果免疫荧光实验结果显示6 h组、12 h组hPDLFs骨架蛋白表达明显高于空白对照组;Western Blot实验结果显示6 h组、12 h组BMP-2蛋白表达明显高于空白对照组且具有统计学意义(P<0.05);ALP染色实验结果显示6 h组、12 h组中ALP活性明显高于空白对照组;茜素红染色实验结果显示6 h组、12 h组骨钙结节形成明显高于空白对照组。结论周期性牵张力的施加会促进hPDLFs中BMP-2的表达并促进hPDLFs成骨分化,可为临床口腔正畸进一步提供理论依据。Objective To investigate the effect of cyclic tensile force on the expression of bone morphogenetic protein-2(BMP-2)in human periodontal ligament fibroblasts(hPDLFs)and its impact on the osteogenic differentiation of hPDLFs.Methods The hPDLFs were cultured in vitro and divided into three groups(blank control,6 h group and 12 h group)with no power-up and power-up for 6 and 12 hours,respectively.Immunofluorescence assay was used to detect the expression of cytoskeletal proteins in the three groups;Western Blot assay was used to detect the expression of BMP-2 in the three groups;the alkaline phosphatase staining kit and the alizarin red staining test were used to detect alkaline phosphatase(ALP)activity and bone calcium nodule formation in three separate groups.Results The results of the immunofluorescence assay showed that the expression of hPDLFs backbone protein was significantly higher in the 6 h and 12 h groups than that in the blank control group;the results of the Western Blot assay showed that the expression of BMP-2 protein was significantly higher in the 6 h and 12 h groups than that in the blank control group and was statistically significant(P<0.05);the results of the ALP staining experiment showed that the ALP activity in the 6 h and 12 h groups was significantly higher than that in the blank control group;the results of alizarin red staining showed that the formation of bone calcium nodules in the 6 h and 12 h groups was significantly higher than that in the blank control group.Conclusion Applying cyclic retraction force promotes the expression of BMP-2 in hPDLFs and the osteogenic differentiation of hPDLFs,which may provide a further theoretical basis for clinical orthodontics.
